Effects of Choulingdan extract modulating TGF-β/BMPs signaling pathway on infected rats after fixation of femoral intertrochanteric fracture
OBJECTIVE To analyze the effect of Choulingdan extract modulating TGF-β/BMPs signaling pathway on infected rats after fixation of femoral intertrochanteric fracture.METHODS A total of 44 SPF Wistar rats were selected,10 rats were used as the blank group,and the remaining 34 rats were used to establish the infection mod-el after fixation of intertrochanteric fracture of the femur.A total of 30 rats were modeled successfully,which were divided into 3 groups,including model group,drug control group and Kuolingdan extract group,with 10 rats in each group.The drug control group was given amoxicillin,the Choulingdan extract group was given Chouling-dan extract,the blank group and the model group were not treated,and the changes of the rats were observed af-ter intervention for 7 days.RESULTS Compared with the blank group,the levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and carboxy-terminal peptide β(β-CTX)of type Ⅰ collagen were increased in model group,drug control group and Choulingtang extract group,while the levels of interleukin-10(1L-10),typeⅠ procollagen N-terminal propeptide(PⅠNP),bone alkaline phosphatase(BALP),transforming growth factor beta(TGF-β)mRNA and Bone morphogenetic protein(BMP-2)mRNA expression and their relative expression levels were de-creased(P<0.05).Compared with model group,the levels of TNF-a,IL-6 and β-CTX in drug control group and Kuol-ingdan extract group were decreased,while the mRNA expressions and relative expressions of IL-10,PINP,BALP,TGF-β mRNA and BMP-2 were increased(P<0.05).Compared with drug control group,the levels of TNF-α,IL-6 and β-CTX were decreased,while the mRNA expression levels of IL-10,PⅠNP,BALP,TGF-β mRNA and BMP-2 mR-NA were increased(P<0.05).CONCLUSION The inflammatory response was alleviated and bone metabolism was im-proved in infected rats after fixation of intertrochanteric fracture of femur by intervention with the extract of Choulingdan,and the mechanism might be related to the activation of TGF-β/BMPs signaling pathway.
