Intervention effect and mechanisms of forsythia extracts on rats with Streptococcus pneumoniae infection-induced acute exacerbation of chronic obstructive pulmonary disease
OBJECTIVE To explore the intervention mechanisms of forsythia extracts on rats with Streptococcus pneumoniae infection-induced acute exacerbation of chronic obstructive pulmonary disease(AECOPD).METHODS Totally 40 SD rats of SPF grade were randomly divided into the normal group,the model group,the low-dose forsythia extract group and the high-dose forsythia extract group,with 10 rats in each group.S.pneu-moniae was chosen to establish the S.pneumoniae infection-induced AECOPD rat models,the control group was not given any treatment.The intervention was carried out after the modeling was done,the ratio of tube wall area to total vessel area(WA%)and the ratio of muscular artery wall thickness to outer diameter of vessel(WT%)were measured.The levels of pulmonary function indexes and inflammatory factors as well as the relative expres-sion levels of mucoprotein 5AC(MUC5AC),epidermal growth factor receptor(EGFR)and phosphorylation-mito-gen-activated protein kinase p38 antibody(p38MAPK)in lung tissues were observed.RESULTS The WT%,WA%,tumor necrosis factor-α(TNF-α)level,interleukin(IL)-6 level,IL-8 level,and the expression levels of p38MAPK,MUC5AC and EGFR were higher in the model group,the low-dose forsythia extract group and the high-dose forsythia extract group than in the control group;the peak expiratory flow(PEF),forced vital capacity(FVC)and forced expiratory volume in one second(FEV1)were lower in the model group,the low-dose forsythia extract group and the high-dose forsythia extract group than in the control group(P<0.05).The expression levels of PEF,FEV1 and FVC of the low-dose forsythia extract group and the high-dose forsythia extract group were higher than those of the model group;while the WA%,WT%,IL-6 level,IL-8 level,TNF-α level,MUC5AC level,p38MAPK level and EGFR level of the low-dose forsythia extract group and the high-dose forsythia extract group were lower than those of the model group(P<0.05);the expression levels of PEF,FVC and FEV1 of the high-dose forsythia extract group were higher than those of the low-dose forsythia extract group,while the WA%,WT%,TNF-α level,IL-8 level,IL-6 level,EGFR level,MUC5AC level and p38MAPK level of the high-dose forsythia extract group were lower than those of the low-dose forsythia extract group(P<0.05).CONCLUSION The forsythia extracts may effectively improve the pulmonary function of the rats with S.pneumo-iae infection-induced AECOPD,inhibit the expression of inflammatory factors,regulate the vascular remodeling of the lungs and improve the prognosis.
