Effect of Hydnoglossus alba extract on regulation of PI3K/AKT/mTOR signaling pathways of hepatocellular carcinoma rats with postoperative infection
OBJECTIVE To observe the effect of Hydnoglossus alba extract on regulation of phosphatidylinositol 3 kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR)signaling pathways of hepato-cellular carcinoma rats with postoperative infection.METHODS Totally 44 Wistar rats of SPF grade were enrolled in the study,10 of which were assigned as the bland group,the rest of 34 were chosen to build up the models of postoperative infection.A total of 30 rats were successfully built up for the models and were divided into the model group,the drug control group and the Hedyotis alba extract group,with 10 rats in each group.The drug control group was treated with amoxicillin and clavulanate potassium,the Hedyotis alba extract group was given Hydno-glossum extract;the blank group and the model group were not given any treatment.The changes of the rats were observed after the interventions were taken for 7 days.RESULTS The levels of C-reactive protein(CRP),interleu-kin-8(IL-8),tumor necrosis factor-α(TNF-α),alanine aminotransferase(ALT),total bilirubin(TBiL)and total bile acid(TBA)as well as the mRNA expression levels and relative expression levels of PI3K,AKT and mTOR were significantly higher in the model group,the drug control group and the Hedyotis alba extract group than in the blank group.(P<0.05).The levels of CRP,IL-8,TNF-α,ALT,TBil,AST and TBA as well as the mRNA expression levels and relative expression levels of PI3K,AKT and mTOR were significantly lower in the drug con-trol group and the Hedyotis alba extract group than in the model group(P<0.05).The levels of CRP,IL-8,TNF-α,ALT,TBil,AST and TBA as well as the mRNA expression levels and relative expression levels of PI3K,AKT and mTOR were significantly lower in the Hedyotis alba extract group than in the drug control group(P<0.05).CONCLUSION Hydnoglossus alba extract may alleviate the inflammatory reactions of the hepatocellular carcinoma rats with postoperative infection and relieve the liver damage,and the mechanisms may be associated with the inhibition of PI3K/AKT/mTOR signaling pathways.
