头花蓼提取物调控PI3K/Akt信号通路对大肠埃希菌感染肾盂肾炎大鼠的干预效果

目的 分析头花蓼提取物通过调控磷脂酰肌醇3激酶/蛋白激酶B(PI3K/Akt)信号通路对大肠埃希菌感染肾盂肾炎大鼠的干预效果.方法 选取SPF级Wistar大鼠45只,10只为对照组,其余35只建立大肠埃希菌感染肾盂肾炎模型,共有30只大鼠成功建模,将其分为模型组、药物对照组、头花蓼提取物组3组,每组10只.药物对照组给予左氧氟沙星灌胃;头花蓼提取物组给予头花蓼提取物灌胃;对照组、模型组不做处理,给予0.5ml生理盐水灌胃,7 d干预后观察各组大鼠的情况变化.结果 与对照组比较,模型组、药物对照组、头花蓼提取物组的白细胞介素(IL)-6、IL-4、IL-10、分泌型免疫球蛋白A(SIg A)、血清肌酐(Scr)、尿素氮(BUN)、24 h蛋白尿量、PI3K、AKT表达量上升(P<0.05);与模型组比较,药物对照组、头花蓼提取物组的SIg A、IL-4、IL-6、IL-10、BUN、24 h蛋白尿量、Scr、PI3K、AKT表达量下降(P<0.05);与药物对照组比较,头花蓼提取物组的IL-6、IL-10、IL-4、SIg A、BUN、Scr、24h蛋白尿量、AKT、PI3K表达量降低(P<0.05).结论 对大肠埃希菌感染肾盂肾炎大鼠给予头花蓼提取物进行干预,可有效降低炎症反应,促进免疫系统对SIg A的分泌,保护大鼠的肾脏组织,以减轻肾损伤,其机制可能与PI3K/Akt信号通路相关.
Effect of Polygonum capulosus extract on rats infected with Escherichia coli pyelonephritis by regulating PI3K/Akt signaling pathway
OBJECTIVE To analyze the intervention effect of Polygonum capulosus extract on Escherichia coli pye-lonephritis rats by regulating phosphatidylinositol 3 kinase/protein kinase B(PI3K/Akt)signaling pathway.METHODS Forty-five SPF Wistar rats were selected and 10 rats were in the control group and the remaining 35 rats were used to establish the model of pyelonephritis infected with E.coli.A total of 30 rats were successfully established as the model.They were divided into three groups including the model group,the drug control group,and the Persicaria capitata extract group,with 10 rats in each group.The control group was given levofloxacin in-tragastric administration.Polygonum capitatum extract group was given by intragastric administration.The control group and model group were given 0.5 ml normal saline intragastric administration without treatment,and the changes of the rats in each group were observed after intervention for 7 days.RESULTS Compared with the control group,Interleukin(IL)-6,IL-4,IL-10,secretory immunoglobulin A(SIgA),serum creatinine(Scr),Blood urea nitrogen(BUN),24-hour Proteinuria,PI3K,AKT expression in the model group,drug control group,and Per-sicaria capitata extract group increased(P<0.05).Compared with model group,SIg A,IL-4,IL-6,IL-10,BUN,24h proteinuria,Scr,P13K and AKT expression levels in the drug control group and Polygonum capuifoli-um extract group were decreased(P<0.05).Compared with the drug control group,the expression levels of IL-6,IL-10,IL-4,SIg A,BUN,Scr,24h proteinuria,AKT and PI3K in the Polygonum capitatum extract group were decreased(P<0.05).CONCLUSION The intervention of Polygonum capuiflorum extract in E.coli infected pyelonephritis rats can effectively reduce the inflammatory response,promote the secretion of SIg A by the im-mune system,protect the kidney tissue of rats,and slow down the kidney injury.
