Effect of Staphylococcus aureus infection on tryptophan metabolism in patients with atopic dermatitis
OBJECTIVE To investigate the effects of Staphylococcus aureus infection on tryptophan metabolism in patients with atopic dermatitis.METHODS A total of 106 patients with atopic dermatitis and 89 healthy volunteers admitted to the 971st Hospital of the Chinese People's Liberation Army Navy from Jan 2020 to Jan 2023 were re-cruited.The skin samples of atopic dermatitis patients were collected. The pathological samples were in the patho-logical group and the non-pathological samples were in the non-pathological group.Skin samples from healthy sub-jects were the control group.Staphylococcus aureus load, proportions of Staphylococcus aureus CC1 strain and CC30 strain, physical and chemical properties of skin barrier, level of tryptophan metabolates, including indole-3-acetic acid(IA),indole-acetic acid(IAA),and indole-in 3 groups of samples were detected 3-lactate(ILA),in-dole-3-propionic acid(IPA),indole-3-aldehyde(I3C),indole-3-formaldehyde(IAId),L-tryptophan(Trp),5-hydroxytryptophan(5-HTP)and L-kynurenine(KYN)].RESULTS Compared with the control group and the non-pathological group, the load of Staphylococcus aureus and the proportion of CC1 strain in the pathological group were higher, while the proportion of CC30 strain was lower(P<0.05);the physicochemical properties of the skin barrier were decreased(P<0.05);the levels of tryptophan metabolites IA and IAA were increased(P<0.05).The levels of tryptophan metabolites such as ILA,IPA and IAId were decreased(P<0.05).The load of Staphylococcus aureus, the proportions of CC1 and CC30 were correlated with the physicochemical properties of the skin barrier.Staphylococcus aureus load, proportions of CC1 strain and CC30 strain were correlated with tryp-tophan metabolites, including IA,IAA,ILA,IPA and IAId.The physical and chemical properties of skin barrier were correlated with tryptophan metabolism products of IA,IAA,ILA,IPA and IAId.CONCLUSION The load of Staphylococcus aureus in the skin of patients with specific dermatitis is higher, and the level of Staphylococcus aureus is correlated with the physicochemical properties of the skin barrier, tryptophan metabolites IA,IAA,ILA,IPA and IAId.
