Correlation of Porphyromonas gingivalis infection with NLRP-3 inflammatory body pathway,HMGB1 and MMP-3 in patients with chronic periodontal disease
OBJECTIVE To investigate the correlation of porphyromonas gingivalis infection with nod like receptor protein-3(NLRP-3)inflammatory body pathway,high-mobility group protein 1(HMGB1)and matrix metallo-proteinase-3(MMP-3)in patients with chronic periodontal disease.METHODS The clinical data of 103 patients with chronic periodontitis admitted to Hengshui city people's hospital from Jan 2021 to Mar 2023 were retrospec-tively collected as the case group.The patients were divided into the infection group(25 cases)and the non-infec-tion group(78 cases)according to whether they were infected with Porphyromonas gingivalis,and they were al-so divided into the mild group(43 cases),the moderate group(32 cases)and the severe group(28 cases)accord-ing to the severity of the disease.The clinical data of 115 periodontally healthy people were collected as the control group.The positive rate of P.gingivalis infection between the case group and the control group was com-pared.The levels of periodontal ligament tissue NLRP-3 inflammatory pathway related indicators and serum HMGB1 and MMP-3 between the case group and the control group,among the chronic periodontitis patients with different severity,and between the infected group and the non-infected group were all compared.The correlation of P.gingivalis infection with periodontal ligament tissue NLRP-3 inflammatory pathway related indicators and ser-um HMGB1 and MMP-3 levels were analyzed by Pearson correlation.RESULTS The positive rate of P.gingival-is infection was 24.27%in the case group,which was higher than that in the control group(11.30%)(P<0.05).The messenger ribonucleic acid(mRNA)levels of periodontal ligament tissue NLRP-3,Caspase-1 and ASC and serum HMGB1 and MMP-3 in the cases group were higher than those in the control group(P<0.05).The levels of periodontal ligament tissue NLRP-3 mRNA,Caspase-1 mRNA,ASC mRNA and serum HMGB1 and MMP-3 in the severe group were higher than those in the moderate and the mild group(P<0.05);the above in-dexes in the moderate group were higher than those in the mild group(P<0.05).The levels of periodontal liga-ment tissue NLRP-3 mRNA,Caspase-1 mRNA,ASC mRNA and serum HMGB1 and MMP-3 in the infected group were significantly higher than those in the non-infected group(P<0.05).Pearson correlation analysis showed that P.gingivalis infection was positively correlated with levels of periodontal ligament tissue NLRP-3 mRNA,Caspase-1 mRNA,ASC mRNA and serum HMGB1 and MMP-3(P<0.05).CONCLUSION The ex-pression levels of periodontal ligament tissue NLRP-3 mRNA,Caspase-1 mRNA,ASC mRNA and serum HMGB1 and MMP-3 were higher in patients with chronic periodontitis,and they were involved in the process of disease progression and closely related to P.gingivalis infection.
