儿童难治性肺炎支原体肺炎RANTES、TIM-3、HMGB1和miR-1323表达水平与病情的相关性

目的 探讨难治性肺炎支原体肺炎患儿外周血微小核糖核酸-1323(miR-1323)、调节活化正常T细胞表达和分泌的趋化因子(RANTES)、T细胞免疫球蛋白黏蛋白分子3(TIM-3)和高迁移率族蛋白B1(HMGB1)水平及其与病情严重程度的关系.方法 选取2020年5月-2023年5月海南医学院第二附属医院收治的97例难治性肺炎支原体肺炎患儿为研究组,同期103名健康体检儿童为对照组,分别采用酶联免疫吸附试验(ELISA)和实时荧光定量聚合酶链式反应(RT-qPCR)检测外周血RANTES、TIM-3、HMGB1和miR-1323表达水平.结果 研究组RANTES、TIM-3、HMGB1和miR-1323水平均高于对照组(P<0.05);难治性肺炎支原体肺炎患儿中度组和重度组RANTES、TIM-3、HMGB1和miR-1323水平高于轻度组(P<0.05),重度组高于中度组(P<0.05);Spearman相关性分析结果显示,RANTES、TIM-3、HMGB1和miR-1323与难治性肺炎支原体肺炎患儿病情严重程度呈正相关(P<0.05).结论 难治性肺炎支原体肺炎RANTES、TIM-3、HMGB1、和miR-1323呈高表达,且与患儿病情严重程度密切相关.
RANTES,TIM-3,HMGB1 and miR-1323 of children with refractory Mycoplasma pneumoniae pneumonia and their association with illness condition
OBJECTIVE To explore the levels of peripheral blood micro ribonucleic acid(miR)-1323,regulate and activate normal T cell expression and secretion factors(RANTES),T cell immunoglobulin mucin molecule 3(TIM-3)and high mobility group protein B1(HMGB1)of the children with refractory Mycoplasma pneumoniae pneumonia and analyze the association with the severity of disease.METHODS A total of 97 children with refracto-ry M.pneumoniae pneumonia who were treated in the Second Affiliated Hospital of Hainan Medical College from May 2020 to May 2023 were assigned as the study group,meanwhile,103 healthy children who received physical examination were chosen as the control group.The expression levels of peripheral blood RANTES,TIM-3,HMGB1 and miR-1323 were detected by respectively using enzyme-linked immunosorbent assay(ELISA)and re-al-time fluorescence quantitative polymerase chain reaction(RT-qPCR).RESULTS The levels of RANTES,TIM-3,HMGB1 and miR-1323 of the study group were significantly higher than those of the control group(P<0.05).Among the children with refractory M.pneumoniae pneumonia,the levels of RANTES,TIM-3,HMGB1 and miR-1323 of the moderate group and the severe group were significantly higher than those of the mild group(P<0.05),the levels of above indexes of the severe group were significantly higher than those of the moderate group(P<0.05).The result of Spearman correlation analysis showed that the RANTES,TIM-3,HMGB1 and miR-1323 were positively correlated with the severity of disease of the children with refractory M.pneumoniae pneumo-nia(P<0.05).CONCLUSION The children with refractory M.pneumoniae pneumonia show the high expressions of RANTES,TIM-3,HMGB1 and miR-1323,which are closely associated with the severity of disease.
