Effect of luteolin on the urinary epithelial barrier in a rat model of recurrent urinary tract infections
OBJECTIVE To investigate the effects of luteolin on the urothelial epithelial barrier in a rat model of re-current urinary tract infection.METHODS A total of 45 SPF SD female rats were selected,including blank group,model group,low dose group and high dose group,with 10 rats in each group.The blank group and model group were only given normal saline intragastric administration without any other treatment,while the low-dose group was given 10 mg/kg luteolin intragastric administration on the basis of the model group,and the high-dose group was given 40 mg/kg intragastric administration.Urine volume,urinary white blood cell levels,inflammatory fac-tors,secreted immunoglobulin A(SIgA),urinary acetylglucosaminase(NAG)levels,urinary epithelial barrier function and the relative expression of toll-like receptor 4/nuclear transcription factor-KB(TLR4/NF-κB)signaling pathway were measured.RESULTS Compared with the blank group,urine volume,E-cadherin and ZO-1 levels in model group,low dose group and high dose group were decreased,while the levels of leukocyte,interleukin-6(IL-6),IL-8,SIgA,NAG,TLR4,NF-κB relative expression were increased(P<0.05).Compared with the model group,urine volume,E-cadherin and ZO-1 levels were increased in the low-dose and high-dose groups,and the relative expression levels of white blood cells,IL-6,IL-8,SIgA,NAG,TLR4 and NF-κB were decreased(P<0.05).Compared with the low dose group,urine volume,E-cadherin and ZO-1 levels were increased in the high dose group,and the levels of white blood cells,IL-6,IL-8,SIgA,NAG,TLR4 and NF-κB relative expres-sion levels were decreased(P<0.05).CONCLUSION Luteolin can improve the urothelial epithelial barrier condi-tion in rats with recurrent urinary tract infection,reduce inflammation,improve voiding function,and regulate immune function,which can reflect the intervention effect on recurrent urinary tract infection by regulating TLR4/NF-κB signaling pathway.
