Intervention effects of honeysuckle extract on post-operative infected rats with endometrial cancer based on H19/miR-195-5p/SGK1 pathway analysis
OBJECTIVE To analyze the intervention effect of honeysuckle extract on post-operative infected rats with endometrial cancer based on H19/MicrornA-195-5P(miR-195-5p)/glucocorticoid-regulated kinase 1(SGK1)pathway.METHODS Totally 70 SPF grade female Wistar rats were selected,and 30 of them were selected for LD35 test.After determining the lethal dose,a dose of 80 mg/kg was selected for administration and observation.10 out of the remaining 40 rats were used as the blank group,30 of them were used to establish the postoperative infection model of endometrial cancer,and a total of 26 rats were successfully modeled,and they were divided into 8 in the model group,9 in the drug-control group,and 9 in the honeysuckle extract group.Drug control group was given amoxicillin and clavulanate potassium,honeysuckle extract group was given honeysuckle extract,blank group and model group were not treated,and the changes of rats were observed after intervention for 14 days.The levels of interleukin-8(IL-8),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),Staphylococcus aureus colony number and the relative expression of H19,micrornA-195-5p(miR-195-5p),and glucocorticoid-regulated kinase 1(SGK1)mRNA expression in each group were detected.RESULTS Compared with the blank group,IL-8,TNF-α,IL-1β,Staphylococcus aureus colony number,H19 and SGK1 mRNA expression levels and relative ex-pression levels were increased in model group,drug control group and honeysuckle extract group,while miR-195-5p mRNA expression levels and relative expression levels were decreased(P<0.05).Compared with model group,interleukin-8(IL-8),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),Staphylococcus aureus colony number,H19 and SGK1 mRNA expression levels and relative expression levels in drug control group and honeylonicera extract group decreased.The mRNA expression and relative expression of miR-195-5p were in-creased(P<0.05).Compared with drug control group,IL-8,TNF-α,IL-1β,Staphylococcus aureus colony num-ber,H19,SGK1 mRNA expression levels and relative expression levels were decreased in honeysuckle extract group,while miR-195-5p mRNA expression levels and relative expression levels were increased(P<0.05).CONCLUSION Honeysuckle extract can reduce the inflammatory response and the number of Staphylococcus au-reus colonies in infected rats with endometrial cancer after operation,and the mechanism may be related to the regulation of H19/miR-195-5p/SGK1.
