Expressions of Th1/Th2 cytokines,miR-122 and miR-155 in peripheral blood of patients with hepatitis B virus infection-associated liver disease and their significance
OBJECTIVE To explore the expressions of helper T cell(Th)1/Th2 cytokines,microRNA(miR)-122 and miR-155 in peripheral blood of the patients with hepatitis B virus(HBV)infection-associated liver disease and analyze the significance.METHODS Totally 102 patients with HBV infection-associated liver disease who were treated in Hai'an Traditional Chinese Medicine Hospital from Jan 2020 to Mar 2023 were recruited as the research subjects and were divided into the chronic hepatitis B group with 45 cases,the hepatitis B liver cirrhosis group with 36 cases,the hepatitis B liver cancer group with 21 cases.Meanwhile,40 healthy people who received physi-cal examination were chosen as the control group.The expression levels of serum miR-122 and miR-155 were de-tected by using real-time fluorescent polymerase chain reaction;the Th1,Th2 and Th1/Th2 were detected by flow cytometry,the liver function indexes were determined by electrochemical luminescence method and automatic im-munoanalyzer.The detection results were compared among the groups.Spearman correlation analysis was per-formed for the association between the serum Th1/Th2 cytokines,miR-122 and miR-155 and the severity of ill-ness condition.RESULTS There were significant differences in the levels of peripheral blood Th1,Th2,Th1/Th2,miR-122,miR-155 and liver function indexes among the four groups(P<0.05).The levels of Th1 and Th1/Th2 of the hepatitis B liver cirrhosis group and the hepatitis B liver cancer group were significantly lower than those of the chronic hepatitis B group,while the levels of Th2,miR-122 and miR-155 of the hepatitis B liver cirrhosis group and the hepatitis B liver cancer group were significantly higher than those of the chronic hepatitis B group;the levels of Th1 and Th1/Th2 of the hepatitis B liver cancer group were the lowest,and the levels of Th2,miR-122 and miR-155 were the highest(P<0.05).The result of Spearman correlation analysis showed that the serum Th1 and Th1/Th2 of the patients with HBV infection-associated liver disease were negatively correlated with the severity of illness condition,while the Th2,miR-122 and miR-155 were positively correlated with the severity of illness condition(P<0.05).CONCLUSION The patients with HBV infection-associated liver disease show high ex-pression levels of serum Th1/Th2 cytokines,miR-122 and miR-155,which are associated with the severity of ill-ness condition.
