Expressions of miR-22 and NLRP3 inflammasomes in peripheral blood mononuclear cells of gastric cancer patients complicated with Hp infection and influence on prognosis
OBJECTIVE To explore the expressions of microRNA-22(miR-22)and NOD-like receptor protein 3(NLRP3)inflammasomes in peripheral blood mononuclear cells(PBMCs)of gastric cancer patients complicated with Helicobacter pylori(Hp)infection and observe the influence on prognosis.METHODS Totally 86 gastric cancer patients complicated with Hp infection who were treated in Wenzhou Integrated Traditional Chinese and Western Medicine Hospital from Jun 2019 to Jun 2022 were assigned as the Hp-positive group,meanwhile,63 gastric cancer patients without Hp infection were chosen as the Hp-negative group.The levels of miR-22,NL-RP3,apoptosis associated speck-like protein containing(ASC)and cysteine aspartate specific proteases-1(Caspase-1)were detected for all of the patients.A 1-year follow-up was conducted for the patients of the Hp-pos-itive group,the patients of the Hp-positive group were divided into the survival group with 72 cases and the death group with 14 cases according to the survival status.The clinical data were collected and compared between the two groups of patients,the expression levels of miR-22 and NLRP3 were compared between the two groups.The effect of the expressions of miR-22 and NLRP3 on prognosis of the gastric cancer patients complicated with Hp in-fection was observed by means of Kaplan-Meier method.RESULTS The expression level of miR-22 in PBMCs was lower in the Hp-positive group than in the Hp-negative group,while the expression levels of NLRP3,ASC and Caspase-1 of the Hp-positive group were significantly higher than those of the Hp-negative bacteria(P<0.05).There were significant differences in depth of invasion,lymphatic metastasis,clinical stage,miR-22,NLRP3,ASC and Caspase-1 between the survival group and the death group(P<0.05).Kaplan-Meier analysis showed that the 1-year survival time of the gastric cancer and Hp infection patients with low expression of miR-22 and high ex-pression of NLRP3 was significantly shorter than that of the patients with high expression of miR-22 and low ex-pression of NLRP3(P<0.05).CONCLUSION The gastric cancer patients complicated with Hp infection show ab-normal expressions of miR-22 and NLRP3 inflammasomes,which are closely associated with the prognosis of the patients.The indexes can be used as major indexes for assessment of the prognosis of the patients.
