TAZ基因多态性与幽门螺杆菌感染的关联

目的 探讨含PDZ结合基序的转录共激活因子(TAZ)基因单核苷酸多态性(SNP)rs16861985、rs1055153、rs6783790、rs12490239与幽门螺杆菌感染的关系。方法 选取2008-2020年包头医学院第一附属医院和内蒙古包钢医院正常体检者470例,通过酶联免疫吸附测定法(ELISA)检测样本中幽门螺杆菌的感染情况,其中幽门螺杆菌阳性248例,幽门螺杆菌阴性222例;采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)技术对TAZ rs16861985、rs1055153、rs6783790、rs12490239进行基因分型,采用非条件性Logistic回归法在共显性、显性、隐性、超显性4种遗传模型下分析各SNP与幽门螺杆菌感染的关系,同时分析赤池信息准则(AIC)和贝叶斯信息准则(BIC)来判断最佳遗传模型;采用SHEsis在线软件对rs16861985、rs1055153、rs6783790、rs12490239进行单体型构建并分析单体型与幽门螺杆菌感染的关系;采用广义多因子降维方法(GMDR)分析TAZ基因SNP-SNP在幽门螺杆菌感染中的交互作用。结果 TAZ rs16861985、rs1055153、rs6783790、rs12490239与幽门螺杆菌感染无关联;TAZ rs16861985、rs1055153、rs6783790、rs12490239构建的单体型与幽门螺杆菌感染无关;TAZ rs16861985、rs1055153、rs6783790、rs12490239的四阶交互作用与幽门螺杆菌感染相关(P<0。05)。结论 TAZ基因rs16861985、rs1055153、rs6783790、rs12490239在包头汉族人群幽门螺杆菌感染中不起主要作用;TAZ基因rs16861985、rs1055153、rs6783790、rs12490239构建的单体型与幽门螺杆菌感染无关;TAZ基因rs16861985、rs1055153、rs6783790、rs12490239在包头汉族人群幽门螺杆菌感染中存在交互作用。
Correlation between TAZ gene polymorphisms and Helicobacter pylori infection
OBJECTIVE To study the associations of the single nucleotide polymorphisms(SNPs)of rs16861985,rs1055153,rs6783790 and rs12490239 in TAZ(transcriptional coactivator containing PDZ binding motif)gene with Helicobacter pylori(Hp)infection.METHODS A total of 470 blood samples of volunteers under normal physical examination in the First Affiliated Hospital of Baotou Medical College and Inner Mongolia Baotou Steel Hospital from 2008 to 2020 were collected.The infection of Hp by enzyme linked immunosorbent assay(ELISA)was detected,including 248 positive cases of Hp and 222 cases of negative for Hp.TAZ rs16861985,rs1055153,rs6783790,rs12490239 were genotyped by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Unconditional logistic regression was used to evaluate the associations of SNPs with Hp infection under codominant,dominant,recessive and overdominant genetic models.And the akaike information criterion(AIC)and bayesian information criterion(BIC)were used to determine the best genetic model.SHEsis online software was used to construct the haplotypes of TAZ rs16861985,rs1055153,rs6783790 and rs12490239,and to analyze the associations of haplotypes with Hp infection.Generalized multifactor dimensionality reduction(GM-DR)was used to analyze the interactions of TAZ gene polymorphisms in Hp infection.RESULTS TAZ rs16861985,rs1055153,rs6783790 and rs12490239 genotypes were not associated with Hp infection.The fourth-order interaction of TAZ rs16861985,rs1055153,rs6783790 and rs12490239 was associated with Hp infection(P<0.05).CONCLUSION Polymorphisms of TAZ gene rs16861985,rs1055153,rs6783790 and rs12490239 have no major effect on Hp infection in Baotou Han population.The haplotypes constructed by TAZ rs16861985,rs1055153,rs6783790 and rs12490239 are not associated with Hp infection.TAZ gene rs16861985,rs1055153,rs6783790 and rs12490239 show interactive effects in Hp infection in Baotou Han population.
