Change of function and polarization levels of macrophages in patients with Chlamydia psittaci pneumonia
OBJECTIVE To explore the change of function and polarization levels of macrophages in the patients with Chlamydia psittaci pneumonia.METHODS A total of 32 patients with C.psittaci pneumonia who were trea-ted in Mindong Hospital Affiliated to Fujian Medical University from Mar 2020 to Feb 2023 were assigned as the pneumonia group,meanwhile,30 healthy people who received physical examination were chosen as the healthy group.The percentage of macrophages in deep respiratory tract secretions and pinocytosis function were detected for all the enrolled research subjects.The expressions of surface markers of macrophages CD86 and CD206 were detected by flow cytometry,and the levels of M1 macrophage polarization markers interleikin-1β(IL-1 β),tumor necrosis factor-α(TNF-α),monocyte chemoattractant protein(MCP-1)mRNA and protein activation were also de-tected;the levels of M2 macrophage polarization markers IL-10,transforming growth factor beta(TGF-β)mR-NA and protein activation as well as the levels of autophagy-related markers Beclin-1,LC3-Ⅰ and LC3-Ⅱ mRNA transcription were detected.RESULTS The percentages of white blood cells and macrophages of the pneumonia group were respectively(12.34±1.49)X 105/ml and(64.87±2.79)%,higher than those of the healthy group(all P<0.05),while the OD value of the pneumonia group was lower than that of the healthy group(P<0.05).The expression levels of macrophage surface markers CD86 and CD206 of the pneumonia group were higher than those of the healthy group(all P<0.05).The transcription levels of M1 macrophage polarization markers IL-1β,TNF-α,MCP-1 mRNA and M2 polarization markers IL-10 and TGF-β mRNA as well as the relative expression levels of proteins were elevated(all P<0.05).The transcription levels of autophagy-related receptors Beclin-1,LC3-Ⅰ and LC3-Ⅱ mRNA and the relative expression levels of proteins of the pneumonia group were higher than those of the healthy group(all P<0.05).CONCLUSION C.psittaci may reduce the endocytosis of the macrophages and pro-mote the polarization.The polarization level of the M1 macrophage is higher than that of the M2 macrophage.
