Association of EB virus infection with miR-222-3p and JAK/STAT signaling pathway in peripheral blood of patients with thyroid cancer
OBJECTIVE To analyze the effects of EB virus(EBV)infection on microRNA(miR)-222-3p and Janus kinase/signal transducer and activator of transcription(JAK/STAT)signaling pathway expression in peripheral blood of patients with thyroid cancer.METHODS From Jan.2021 to Jan.2024,a total of 100 patients who were diagnosed as thyroid cancer combined with EBV infection and hospitalized in the Affiliated Hospital of Inner Mon-golia Medical University were selected as the infection group,and further divided into low-load group with 49 ca-ses,moderate-load group with 33 cases and high-load group with 18 cases according to EBV viral load.Another 60 patients with thyroid cancer during the same period were selected as the non-infection group.MiR-222-3p and JAK/STAT signaling pathway expressions in peripheral blood of the two groups were analyzed.The receiver oper-ating characteristic(ROC)curves were plotted to evaluate the diagnostic value of miR-222-3p and JAK/STAT signaling pathway for thyroid cancer combined with EB virus infection.RESULTS The levels of miR-222-3p,p-JAK1/JAK1,p-JAK2/JAK2 and p-STAT1/STAT1 in peripheral blood of the infection group were 0.28±0.10,2.13±0.38,1.94±0.21 and 2.81±0.34,respectively,which were higher than those in the non-infection group(P<0.05).The miR-222-3p,p-JAK1/JAK1,p-JAK2/JAK2 and p-STAT1/STAT1 in the low load group were 0.22±0.08,1.94±0.36,1.80±0.29 and 2.64±0.41,respectively,which were lower than those in the medium and high load groups(P<0.05),and those in the medium load group were also lower than those the high load group(P<0.05).The area under the ROC curve(AUC)of miR-222-3p combined with JAK/STAT signaling pathway was 0.892 in the diagnosis of EB virus infection in patients with thyroid cancer,which was the largest compared to those of single indexes.CONCLUSIONS EB virus infection upregulates miR-222-3p expression in pe-ripheral blood and activates the JAK/STAT signaling pathway,which may be involved in the occurrence and de-velopment of thyroid cancer.Peripheral blood miR-222-3p and JAK/STAT signaling pathway are expected to be biological indicators for diagnosis of EB virus infection of thyroid cancer patients.
