Association of SNP Loci of POU1F1 Gene with Body Mass and Morphological Trait of Oreochromis niloticus
[Objective]The purpose of this study is to analyze the single nucleotide polymorphisms(SNP)of POU1F1 gene and evaluate whether these polymorphisms are related to the body mass and morphological traits of Nile tilapia(Oreochromis niloticus),and provide valuable information for improving the growth characteristics of Nile tilapia.[Method]A total of 28 loci with high polymorphism were screened from POU1F1 using PCR(product sequencing methods).We analyzed association of these loci with mass and morphological traits in the Gaoyao parent population,and validated it in the Gaoyao offspring population and Panyu population.The obtained body mass and morphology related loci were further validated in the Hainan population.[Result and Conclusion]The results showed that there were 6 loci[S3(A-400G),S4(A-469T),S5(I-539D),S6(A-881G),S7(A-888G)and S12(C-1365T)]and 5 loci[S3,S5,S11(I-1358D),S13(C-1511T),S14(A-1539T)]associated with body mass and morphological traits in the Gaoyao parent and offspring populations,respectively.Among the 11 SNP of POU1F1 gene,no locus associated with body mass and morphological traits in Panyu population were found.The association analysis of six SNP loci of POU1F1 gene with Hainan female and male populations showed that S4 loci were related to the body mass of Hainan female populations,and S3 and S5 loci were related to the body mass of male populations.The correlation analysis between the diplotype and the body mass and morphology traits of each population showed that two diplotypes related to body thickness were obtained in the Gaoyao parent population;In Gaoyao progeny population,Panyu population and Hainan male population,no diplotype related to body mass and morphologicl traits was obtained;One diplotype related to body mass was obtained in Hainan female population.
