Effects of Acute Hypoxic Stress on Expression of Growth-,Immunity-,and Biomineralization-related Genes of Pinctada fucata martensii
[Objective]To investigate the effects of acute hypoxic stress on the expression of growth-,immunity-and of biomineralization-related genes of Pinctada fucata martensii,and to provide a reference for the selection and breeding of stress-resistant strains of P.fucata martensii.[Method]The expression levels of immune and antioxidant activity-related genes(caspase-3,HSP90,CAT,and SOD),growth-related genes(EGFR,FGF18,OSR1,and tgfbr1),as well as biomineralization-related genes(pif177,aspein,nacrein,and TIMP)of P.fucata martensii at time points of 0,12,24,and 48 hours under hypoxic stress were compared,when the pearl oyster was moved from normoxic conditions to a water body with low dissolved oxygen at 2 mg·L-1.[Result]After acute hypoxic stress,the expression level of HSP90 was lower than that of the control group at 12 h(P<0.05),and the expression levels of caspase-3 and CAT were significantly increased at 24 h of stress(P<0.05),whereas there was no significant change in the expression of SOD(P>0.05);The expression of biomineralization-related genes pif177,aspein,and nacrein were all significantly reduced after acute hypoxic stress,and TIMP was significantly reduced only at 12 h(P<0.05).The expression of growth-related genes EGFR,FGF18,and OSR1 genes also showed a significant decrease(P<0.05),and the expression of tgfbr1 was significantly higher than that of the control group at 24 h(P<0.05).[Conclusion]Acute hypoxic stress up-regulated the expression of caspase-3 and CAT but suppressed the expression of HSP90 of immune and antioxidant-related genes,suppressed the expression of biomineralization-related genes,suppressed the expression of EGFR,FGF18,and OSR1 but up-regulated the expression of tgfbr1 of growth-related genes.
