Cultivation Optimization and Algicidal Substances Characteristics of an Algicidal Bacterium on Skeletonema costatum
Harmful red tide is a harmful marine ecological phenomenon,and Skeletonema costatum is one of the algae that cause harmful algae blooms.In the previous study,a strain of algicidal bacteria was screened,which was a species of Bacillus(Bacillus sp.2-1-2),it has obvious inhibitory effect on the S.costatum,but the content of this algicidal bacteria is very small in natural water,and the specific inhibition mechanism has not been clear.To explore the optimal cultivation conditions,stability and composition of algicidal bacteria,and to lay a foundation for the actual water application of algicidal bacteria.First,a single factor test was used to ana-lyze the effects of initial pH,temperature and salinity on the growth and effect of algicidal bacteria,and the optimal culture conditions were obtained.Secondly,single-factor test and orthogonal test were used to analyze the effects of pH,temperature,salinity and ultraviolet light on the stability of algicidal substances.Finally,the possible components of algicidal substances were preliminarily explored by enzymatic hydrolysis test and HPLC.pH 8.0,temperature 25 ℃,salinity 34 were the optimal culture conditions for algicidal bacteria within the experimental conditions.Algicidal substances have the characteristics of acid and alkali resistance,high temperature resistance,high salt resistance,and ultraviolet resistance,and are most affected by pH,followed by salinity and temperature,and finally ultraviolet light.It is speculated that the algicidal substance may be a non-protein substance with high temperature resistance and high algicidal activity under alkaline conditions,and the algicidal activity may be related to RNA.By improving the culture conditions,the biomass of algicidal bacteria can be amplified,and the algicidal substances have certain tolerance to the environment and their components are related to RNA,which provides a new possibility for the active components of algicidal sub-stances.
