大黄鱼氧依赖信号通路关键基因的鉴定与表达分析

鉴定了大黄鱼EGLN家族的EGLN1、EGLN2、EGLN3和HIF1α基因.发现这4个基因所编码的蛋白都是膜外蛋白,没有跨膜区.P4Hc的结构域为EGLN家族的特征结构域.多序列比对发现了这4个基因的保守性.蛋白—蛋白相互作用(PPI)网络分析发现,整个EGLN家族与HIF1α、VHL、TCEB1和EPAS1直接相互作用.系统发育分析验证了 EGLN1在耐受低氧中起关键作用.实时荧光定量PCR分析显示这4个基因在所有组织中表达.在缺氧胁迫下,除了鳃和脑组织,EGLN1的表达先下调再上调.这些结果为大黄鱼养殖生物学提供了基础资料.
Characterization and Expression Analysis of Key Genes of the Oxygen-Dependent Signaling Pathway in Larimichthys crocea
In this study,EGLN1,EGLN2,EGLN3 of EGLN family and HIF1α gene of Larimichthys cro-cea were identified.It was found that the proteins encoded by these four genes were all extracellular proteins without transmembrane region.The domain of P4Hc is the characteristic domain of EGLN family.Multiple se-quence alignment revealed conformance of the four genes.Protein-protein interaction(PPI)network analysis re-vealed that the entire EGLN family directly interacts with HIF1α,VHL,TCEB1 and EPAS1.Phylogenetic analysis confirmed that EGLN1 plays a key role in tolerating hypoxia.Real-time quantitative PCR analysis showed that the four genes were expressed in all tissues.Under hypoxic stress,the expression of EGLN1 was first down-regulated and then up-regulated in all tissues except gill and brain.These results provide basic da-ta for culture biology of L.crocea.
