LRP1在消化系统恶性肿瘤侵袭和预后中的调节作用

目的 探讨低密度脂蛋白受体相关蛋白1(LRP1)在消化系统恶性肿瘤发生发展中的作用.方法 应用癌症基因组图谱(TCGA)数据库分析LRP1在肿瘤中的表达,并通过R软件(3.6.3版)分析LRP1与消化系统恶性肿瘤患者的生存及预后间的关系.临床上对胃癌、肝癌和胰腺癌患者的癌旁组织和肿瘤组织取材,进行HE染色观察LRP1表达量.应用RNA干扰慢病毒降低LRP1在消化系统恶性肿瘤细胞中的表达,观察LRP1敲低后消化系统恶性肿瘤细胞的生物学特性.通过Western blot检测LRP1敲低后消化系统恶性肿瘤细胞蛋白表达变化,探索LRP1影响消化系统恶性肿瘤细胞增殖、侵袭和迁移的机制.结果 泛癌分析显示LRP1在胃癌、胰腺癌和肝癌等组织中高表达.生物信息学分析显示,LRP1高表达与消化系统恶性肿瘤的生存期短、预后不良密切相关.组织水平上,与正常组织相比LRP1在胃癌、肝癌和胰腺癌中高表达,与生物信息学分析结果吻合.蛋白水平检测结果显示,LRP1在HGC-27细胞、HepG2细胞和BxPC-3细胞中高表达.且降低LRP1在消化系统恶性肿瘤细胞中的表达可以抑制EGFR-AKT通路,从而影响MMP的表达进一步抑制消化系统恶性肿瘤细胞的增殖、侵袭和迁移.结论 降低LRP1表达可抑制消化系统恶性肿瘤细胞的增殖、侵袭和迁移,提示LRP1可能成为治疗消化系统恶性肿瘤的新靶点.
Objective To detect the role of low-density lipoprotein receptor-related protein 1(LRP1)in malignant tumors of the digestive system.Methods The cancer genome atlas(TCGA)database was used to analyze the expression of LRP1 in tumors.R software(version 3.6.3)was used to analyze the relationship in survival and prognosis between LRP1 and malignant tumors of the digestive system.The paracancerous tissues and tumor tissues of patients with gastric cancer,liver cancer and pancreatic cancer were collected for HE staining to observe the expression of LRP1.LRP1 expression were reduced by using RNA interfere lentivirus in malignant tumor cells of the digestive system,and malignant tumor cells of the digestive system biological characteristics were observed after LRP1 knocked down.Detected changes in protein expression of malignant tumor cells in the digestive system after LRP1 knockdown through Western Blot,explored the mechanisms by which LRP1 influences the proliferation,invasion,and migration of malignant tumor cells in the digestive system.Results Pan-cancer analysis revealed LRP1 was highly expressed in gastric cancer,colorectal cancer,pancreatic cancer and liver cancer tissues.Bioinformatics analysis showed that high expression of LRP1 was closely related to the short survival time and poor prognosis of malignant tumors of the digestive system.At the tissue level,LRP1 was highly expressed in gastric cancer,liver cancer and pancreatic cancer compared with adjacent normal tissues,which was consistent with the results of bioinformatics analysis.The results of the protein levels were shown that LRP1 was highly expressed in HGC-27 cell,HepG2 cell,and BxPC-3 cell.Results showed that reduced LRP1 expression suppressed the proliferation,invasion,and migration of malignant tumor cells in the digestive system.Conclusion Reduce LRP1 expression inhibited the proliferation,invasion,and migration of malignant tumor cells in the digestive system,suggesting that LRP1 may be a new target for the treatment of malignant tumors in the digestive system.
