Objective To exploring the biological characteristics of obese patients with breast cancer based on metabolomics and gut microbiota.Methods Collected the basic information and fecal samples of breast cancer in Zhejiang Hospital of Traditional Chinese Medicine from January 2021 to January 2023.16s RNA high-throughput sequencing and liquid chromatography-mass spectrometry(LC-MS)were used to detect the difference of intestinal flora and intestinal metabolites between healthy controls and breast cancer patients(normal weight,obesity).Then the metabolic pathway related to obesity in breast cancer were explored.Meanwhile combine intestinal microbes with metabonomics to elucidate the correlation between gut microbiota and metabolites.Results There were total of 16 person's fecal and serum samples in the study,including five healthy controls,six normal weight patients with breast cancer,and five obese patients with breast cancer.The results of gut microflora showed that there were eleven different genus between normal weight breast cancer patients and healthy controls,five different genus between obese breast cancer patients and healthy controls,and twenty-two different genus between normal weight breast cancer patients and the obese breast cancer patients.The metabolomics results showed that there were 170 different blood metabolites(42 down and 128 up)between normal weight breast cancer patients and healthy controls(P<0.05),which were mainly enriched in amino acid metabolism,D-glutamine and D-glutamic acid metabolism,sphingomyelin metabolism,pyruvate metabolism,butyric acid metabolism and other pathways(P<0.05).The level of cholic acid were significantly increased and the level of deoxycholic acid significantly decreased in normal weight breast cancer patients.Compared with the healthy control group,there were 130 different blood metabolites(61 down regulated and 69 up regulated)(P<0.05),mainly concentrated in arachidonic acid metabolic pathway.Compared with normal weight breast cancer patients,there were 160 different blood metabolites(97 down regulated and 63 up regulated)(P<0.05),mainly enriched in primary bile acid biosynthesis.Conclusion There are differences in gut microbiota and serum metabolites among breast cancer patients(normal weight,obesity).And the differential serum metabolites are mainly enriched in the primary bile acid metabolism pathway.
