目的 基于GEO数据库获得阻塞性睡眠呼吸暂停低通气综合征(OSAHS)的相关差异表达基因,探讨其分子机制,并进行中药预测,以期为OSAHS的中西医结合诊疗提供理论依据及新的思路。方法 在GEO数据库中以"Obstructivesleepapnea-hypopneasyndrome"为关键词检索,获得芯片数据集GSE135917,采用GEO2R在线分析筛选差异表达基因,通过Excel进行数据整理与分析,获得显著差异基因,对其进行GO和KEGG富集分析、蛋白互作(PPI)网络构建及可视化分析,得到关联的信号通路及核心基因,最后通过Coreminemedical数据库进行核心基因映射,得到与疾病相关中药。结果 OSAHS相关显著差异表达基因共105个;GO富集分析显示差异基因的生物进程主要集中在对脂多糖、糖皮质激素、细菌来源分子、皮质类固醇等反应中,细胞组分主要富集在低密度脂蛋白颗粒、内吞囊泡管腔、高密度脂蛋白颗粒、溶酶体腔等中;分子功能主要以RNA聚合酶Ⅱ特异性DNA结合转录因子结合、生长因子活性、DNA转录因子结合、核受体结合为主;KEGG富集分析共得到10条信号通路,包括IL-17信号通路、TNF信号通路、卡波西肉瘤相关疱疹病毒感染通路、致病性大肠杆菌感染通路等;蛋白互作网络构建及可视化分析共得到12个核心基因(IL-6、FOS、FOSB、NR4A1、ATF3、DUSP1、EGR1、JUN、ZFP36、PTGS2、CXCL2、SOCS3),核心基因映射共获得89味具有潜在治疗效果的中药,功效以补虚药、清热药、活血化瘀药、化痰止咳平喘药、祛风湿药为主。结论 IL-6、FOS等核心基因的表达及IL-17、TNF等信号通路可能在OSAHS病程中起到关键作用,部分清热活血化瘀、补虚化痰止咳的中药可靶向治疗OSAHS,具有潜在治疗效果及良好的应用前景,生物信息学分析可得到OSAHS治疗的多个靶点,对其精准治疗及中西医结合治疗具有重要意义。
Objective To obtain the differentially expressed genes related to obstructive sleep apnea Hypopnea syndrome based on GEO database through bioinformatics technology,to explore its molecular mechanism,and to make predictions on traditional Chinese medicine,in order to provide theoretical basis and new ideas for the integrated diagnosis and treatment of obstructive sleep apnea Hypopnea syndrome.Methods The chip dataset GSE135917 was retrieved by using the keyword"Obstructive sleep apnea homodyne"in the GEO database.GEO2R online analysis was used to screen differentially expressed genes,and the Excel was used to organize and analyze the data to obtain significantly differentially expressed genes.GO and KEGG enrichment analysis,protein interaction(PPI)network construction,and visualization analysis were conducted to obtain the associated signaling pathways and core genes.Finally,the core gene mapping was performed through the Coreminemedical database to obtain disease-related traditional Chinese medicine.Results This study showed a total of 105 differentially expressed genes related to OSAHS.GO enrichment analysis showed that the biological processes of differential genes were mainly concentrated in the reactions to Lipopolysaccharide,glucocorticoid,bacteria derived molecules,Corticosteroid,etc.The cell components were mainly enriched in low-density lipoprotein particles,endocytosis vesicle lumens,High-density lipoprotein particles,lysosome lumens,etc.The main molecular functions are RNA polymerase Ⅱ specific DNA binding transcription factor binding,growth factor activity,DNA transcription factor binding,and Nuclear receptor binding.KEGG enrichment analysis yielded a total of 10 signal pathways,including IL-17 signal pathway,TNF signal pathway,Kaposi's sarcoma associated herpesvirus infection pathway,pathogenic Escherichia coli infection pathway,etc.A total of 12 core genes(IL-6,FOS,F OSB,NR4A1,ATF3,DUSP1,EGR1,JUN,ZFP36,PTGS2,CXCL2,SOCS3)were obtained through protein interaction network construction and visualization analysis.A total of 89 Chinese herbal medicines with potential therapeutic effects were obtained through core gene mapping,with the main effects being tonifying deficiency,clearing heat,promoting blood circulation and resolving stasis,resolving phlegm,cough,and asthma,and expelling wind and dampness.Conclusion The expression of core genes such as IL-6,FOS,and signaling pathways such as IL-17,TNF may play a key role in the course of OSAHS.Some traditional Chinese medicines for clearing heat,promoting blood circulation,removing blood stasis,tonifying deficiency,resolving phlegm,and relieving cough can Targeted therapy OSAHS,with potential therapeutic effects and good application prospects.Bioinformatics analysis can obtain multiple targets for OSAHS treatment,which is of great significance for its precise treatment and integrated traditional Chinese and western medicine treatment.
