Objective To explore the relationship between periodontitis and IL-18 and its polymorphic sites.Methods The plasma IL-18 level of all subjects was measured,and the relationship between plasma IL-18 and periodontitis was analyzed.To compare the correlation between different genotypes and the risk of periodontitis,we sequenced the rs187238 and rs1946518 loci of IL-18 gene in 323 subjects.Results The level of plasma IL-18 expression in patients with periodontitis was 463.06±48.04pg/ml,which was higher than that in the healthy control group,and the difference was statistically significant(P<0.05).Multivariate logistic regression analysis showed that the distribution of rs187238 GC genotype in patients with periodontitis was significantly higher than that in the healthy controls(OR=0.36,95%CI:0.21~0.64,P<0.05).The distribution of AA genotype at rs1946518 locus in patients with periodontitis was also significantly higher than that in the healthy controls(OR=0.53,95%CI:0.29~0.96,P<0.05).The results of hierarchical analysis of rs187238 locus showed that the frequency of heterozygous genotype GC at this locus in patients with periodontitis was significantly higher than that in the healthy controls(P<0.05).The haplotype study showed that individuals carrying AA haplotypes could increase the risk of periodontitis.The combined effect of genes also found that the risk of periodontitis increased with the increase of carrying risk genes.The haplotype study showed that the individuals carrying CA and GA haplotypes in patients with periodontitis were significantly higher than those in the healthy control group(P<0.05),suggesting that carrying these two haplotypes could increase the risk of periodontitis.Conclusion IL-18 and its genetic variation may play an important role in the pathogenesis of periodontitis in Chinese Han population.
