Objective To explore the clinical efficacy and mechanism of action of Banxia Shumi Tang in the treatment of chronic insomnia.Methods Sixty patients with chronic insomnia were enrolled and randomly divided into a treatment group and a control group.The treatment group was treated with Banxia Shumi Tang,while the control group was treated with Zaoren Anshen Granules.The Pittsburgh Sleep Quality Index(PSQI)and Athens Insomnia Scale(AIS)scores of the two groups of patients were evaluated after admission,4 weeks of treatment,and 8 weeks of treatment.Polysomnography and serum Brain-gut peptide,such as SP,NPY and 5-HT,were performed after admission and 4 weeks of treatment.So as to analyze the clinical efficacy and mechanism of action of the two groups.Results At the follow-up of 4 and 8 weeks of treatment,both groups showed significant improvement in PSQI and AIS scores(P<0.01),and the treatment group showed more significant improvement in PSQI and AIS(P<0.05).After 4 weeks of treatment,the total sleep time,sleep latency,number of awakenings,and sleep efficiency of the treatment group were significantly improved compared to before treatment(P<0.01),and were significantly better than the control group in improving sleep latency,number of awakenings,and sleep efficiency(P<0.01).At the same time,the Brain-gut peptide SP,NPY,and 5-HT of patients in the treatment group were significantly improved compared to before treatment(P<0.01),and were significantly better than the control group in improving SP and NPY(P<0.01).In addition,there was a positive linear regression relationship(P<0.05)between PSQI score and SP,and a negative linear regression relationship(P<0.01)with NYP and 5-HT.Conclusion Banxia Shumi Tang can significantly improve the sleep quality of patients with chronic insomnia,while improving the total sleep time,sleep latency,number of awakenings,and sleep efficiency.Its mechanism may be related to regulating the expression of Brain-gut peptides.This research provides new ideas and reliable basis for the clinical prevention and scientific research of insomnia in traditional Chinese medicine.It is worth further research and promotion.
