Objective To investigate the expression,prognosis of SHC SH2-domain binding protein 1(SHCBP1)and its effect on immune cell infiltration in lung adenocarcinoma by bioinformatics methods.Methods The transcriptional,clinical and survival information data of LUAD were downloaded from the Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO)databases,and the difference of SHCBP1 expression between LUAD and normal lung tissues was compared.The relationship between SHCBP1 expression and clinical parameters,clinical prognosis and independent prognostic factors of LUAD were analyzed.The infiltration of immune cells in SHCBP1 high and low expression groups was evaluated.SHCBP1-related signaling pathways were predicted.Results Compared with normal lung epithelial tissues,the expression of SHCBP1 was significantly increased in LUAD tissues.The expression of SHCBP1 is related to age,T N stage and clinical stage of LUAD patients.Compared with the SHCBP1 low expression group,the overall survival(OS),disease specific survival(DSS)and progression-free survival(PFS)of the SHCBP1 high expression group were shorter.Multivariate COX regression analysis showed that SHCBP1 was an independent prognostic factor for LUAD patients.GSEA results showed that the highly expressed genes of SHCBP1 were enriched in the genes of mitotic sister chromatid separation,chromosome separation and sister chromatid separation.CIBERSORT and ssGSEA analysis showed that compared with the low expression group,the number of tumor-infiltrating immune cells such as CD4 and CD8T cells,natural killer T cells,helper T cells and memory B cells in the SHCBP1 high expression group increased.Conclusion SHCBP1 is up-regulated in LUAD tissues,and high expression of SHCBP1 is associated with poor prognosis of LUAD.SHCBP1 can be used as a potential target and prognostic biomarker for immunotherapy of LUAD.
