Objective To explore the effect of Yanluotong on the ET-1/ETAR signaling pathway in a rat retinal vein occlusion(RVO)model.Methods 48 healthy male SD rats were randomly divided into a blank control group(Control),a model control group(Model),a Bosentan group,and a Yanluotong group.Twelve RVO model SD rats were established using photochemical methods and administered continuously for three weeks.At the third week,SD rats were euthanized and serum was collected for Elisa detection.Retinal tissue was taken for fluorescence quantitative PCR and immunofluorescence detection,and the protein and mRNA expression levels of ET-1,ETAR,VEGF,and TNF-α,IL-6 were measured.Results The level of serum IL-6,TNF-α and VEGF in the model group were significantly increased compared to the blank control group(P<0.01),while after intervention with Bosentan and Yanluotong Formula for 3 weeks,VEGF levels were significantly decreased compared to the model group(P<0.01).Immunofluorescence showed that the expression level of ET-1,IL-6,TNF-α and VEGF increased in the RVO model group compared to the normal blank group,but after treatment with Bosentan and Yanluotong Formula,the expression level showed a downward trend.For the mRNA expression levels of ET-1,ETAR,and VEGF in the model control group was significantly upregulated compared to the blank control group(P<0.01),and the use of bosentan and yanluotong formula could significantly downregulate the mRNA expression of these genes(P<0.01).Conclusion The ET-1/ETAR signaling pathway is abnormally activated during the pathogenesis of RVO.Yanluotong can exert inhibitory effects on retinal vasoconstriction,alleviate inflammatory response,inhibit VEGF,and intervene in the progression of RVO through the ET-1/ETAR signaling pathway.
