目的 检测前列腺癌中紧密连接蛋白-4(Claudin-4)和基质金属蛋白酶-9(MMP-9)蛋白的表达,探讨Claudin-4和MMP-9蛋白表达与前列腺癌临床预后的相关性。方法 采用组织微阵列和免疫组化SP法检测85例前列腺癌和41例前列腺增生组织中Claudin-4和MMP-9蛋白表达,分析其与临床病理及5年生化复发率和总体生存率的相关性。结果 前列腺癌中Claudin-4和MMP-9蛋白表达阳性率分别为64。56%和58。23%;前列腺增生中表达阳性率分别为33。33%和20。00%,两者比较差异有统计学意义(P<0。05);Claudin-4和MMP-9蛋白表达与前列腺癌TNM分期、Gleason评分、术前PSA、阳性切缘和5年生化复发率和总体生存率有关(P<0。05)。Pearson相关性分析显示,前列腺癌中Claudin-4和MMP-9蛋白表达呈正相关(P<0。05)。Kaplan-Meier分析显示,Claudin-4和MMP-9蛋白表达增高组前列腺癌5年生化复发和总体生存高于对照组(P<0。05)。结论 Claudin-4和MMP-9蛋白在前列腺癌中呈高表达与前列腺癌的发生和发展有关。
Objective To detect the expression of Claudin-4(Claudin-4)and Matrix metalloproteinase-9(MMP-9)in prostate cancer,and to explore the relationship between the expression of Claudin-4 and MMP-9 in prostate cancer with clinical prognosis of cancer.Methods The expression of Claudin-4 and MMP-9 proteins was detected in 85 cases of prostate cancer and 41 cases of prostate hyperplasia using tissue microarray and immunohistochemical methods,and their correlation with clinical pathology,5-year biochemical recurrence rate,and overall survival rate was analyzed.Results The positive rates of Claudin-4 and MMP-9 protein expression in prostate cancer were 64.56%and 58.23%.The positive rates in benign prostatic hyperplasia were 33.33%and 20.00%,respectively,and there was a significant difference(P<0.05).Claudin-4 and MMP-9 protein expression correlated with prostate cancer TNM staging and Gleason score,preoperative PSA,positive resection margin,5-year biochemical recurrence rate and overall survival rate(P<0.05).Pearson correlation analysis showed a positive correlation between Claudin-4 and MMP-9 protein expression in prostate cancer(P<0.05).Kaplan-Meier analysis showed that the increased expression of Claudin-4 and MMP-9 protein were higher than those in the control group about the 5-year biochemical recurrence and overall survival of prostate cancer(P<0.05).Conclusion The high expression of Claudin-4 and MMP-9 protein in prostate cancer is related to the occurrence and development of prostate cancer.
