Objective To investigate the effect and mechanism of Glaucocalyxin A(GLA)on the biological behavior of osteosarcoma(OS)cells.Methods Different concentrations of GLA were applied to osteosarcoma cells(143B,HOS,U2OS).The CCK8 method was used to detect the cell activity after 24,48 and 72 h of GLA treatment,and the cell line with the most obvious effect was selected as the tool cell line for study.The cell proliferation ability was detected by the cell clone formation assay.The cell migration ability was detected by the scratch assay.The cell invasion ability was detected by the transwell assay.The cell apoptosis was detected by the flow cytometry.RT-qPCR and Western Blot assay were used to investigate the potential molecular mechanism.Results CCK8 assay showed that GLA inhibited the malignant proliferation of osteosarcoma cells in a concentration-dependent manner,and the proliferation of 143B cell line was most significantly inhibited.The cell clone formation assay,scratch assay,Transwell assay and flow cytometry showed that GLA significantly inhibited the proliferation,migration and invasion ability of 143B cells and promoted the apoptosis of 143B cells.RT-qPCR assay and Western Blot assay showed that GLA significantly downregulated the mRNA and protein expression levels of proteins related IL-6/STAT3 pathway,but did not affect the expression of downstream proteins related IL-6/MAPK cascade pathway and IL-6/PI3K pathway.In addition,the Western Blot assay also showed that GLA significantly inhibited cell cycle and apoptosis-related proteins(Cyclin D1,Bcl-2,XIAP).Conclusion GLA may inhibit the proliferation,migration and invasion of osteosarcoma cells and induce apoptosis by regulating the IL-6/STAT3 pathway.
