桃红四物汤治疗增生性瘢痕的作用机制

目的 应用网络药理学筛选桃红四物汤(DSO)中的活性成分,预测其对增生性瘢痕(HS)的作用机制。方法 通过TCMSP、NCBI数据库和PharmMapper数据库预测DSO的潜在靶点,用GeneCards、OMIM、PharmGKB等数据库收集HS的相关靶点。将DSO的潜在靶点与HS的靶点交集,获得共同靶点。通过STRING蛋白数据库并采用Cytoscape 3。8。2绘制DSO-HS的蛋白质-蛋白质相互作用图(PPI),筛选核心靶点。将共同靶点上传至DAVID数据库进行GO和KEGG分析。通过免疫印迹实验(Western blot)验证DSO对筛选出的通路的调控作用。结果 共筛选出DSO潜在靶点425个,HS相关靶点1285个,共同靶点112个,其中核心靶点为ALB、AKT1、MMP-9。通路分析结果显示DSO参与多条相关通路的调控,主要涉及PI3K-Akt信号通路、Ras信号通路、MAPK信号通路等。免疫印迹试验实验结果验证DSO对AKT通路的调控作用。结论 DSO中的多个组分可能通过AKT等多核心靶点,影响多个通路治疗HS。
Objective To screen the active components of Taohong Siwu Decoction(THSWD)using network pharmacology and predict its mechanism of action on hypertrophic scars(HS).Methods The ingredients and the potential targets of THSWD were predicted with TCMSP,NCBI database and PharmMapper database,and the relevant targets of HS were collected by GeneCards,OMIM,PharmGKB and other databases.The both targets'collections were merged to obtain the ingredients-disease targets.And the protein-protein interaction(PPI)network was prepared with STRING protein database by using Cytoscape 3.8.2 to select the highly focused targets.The DAVID database was applied for GO and KEGG pathway analysis.Immunoblotting assay(Western Blot)was performed to verify the regulatory effects of Tao Hong Si Wu Tang on the selected pathway.Results A total of 425 potential targets of THSWD,1285 HS-related targets were selected,112 of which were highly focused.And the top three targets were ALB,AKT1 and MMP-9.The results of the signaling pathway analysis highly lighted PI3K-Akt signaling pathway,Ras signaling pathway and MAPK signaling pathway et al.The Western blot results showed that Tao Hong Si Wu Tang could regulate the AKT pathway.Conclusion Multiple components in DSO may treat HS by targeting multiple pathways through core targets such as AKT.
