炙马钱子和马钱子碱氮氧化物的毒性研究

目的 明确炙马钱子和马钱子碱氮氧化物的毒性。方法 选用C57BL/6小鼠210只,雌雄各半,随机分为对照组10只,生马钱子组、炙马钱子组、马钱子碱组、马钱子碱氮氧化物组,各50只。对照组正常饲养饮水,其他组灌胃给药,观察24 h内小鼠中毒死亡情况,然后持续观察记录14 d内小鼠死亡情况,逐日记录各组小鼠死亡数量和中毒症状。灌胃给药,观察小鼠的一般状态、死亡情况、给药后14 d内的体质量变化;采用HE染色观察死亡小鼠心、肝、脾、肾组织的病理损伤情况。结果 给药后四种药品的小鼠中毒反应类似,并呈现剂量依赖性加重。14 d内各给药组小鼠的体质量变化较对照组无明显异常。炙马钱子的LD50约为生马钱子的1。6倍;马钱子碱氮氧化物的LD50可达马钱子碱的31倍左右。相较于炙马钱子和马钱子碱氮氧化物,生马钱子和马钱子碱的死亡-剂量曲线,较为陡直,均呈剂量依赖性。与对照组相比,四种药品最高剂量组中毒死亡小鼠心、肝、脾、肾组织HE染色均未见明显异常。结论 炙马钱子和马钱子碱氮氧化合物比生马钱子和马钱子碱毒性更小,治疗窗更大,更安全。
Objective To clarify the toxicity of processed Strychnine Semen and Brucine N-oxide,use Strychnine Semen more safely,and further develop and utilize Strychnine Semen and Brucine N-oxide.Methods A total of 210 C57BL/6 mice,half male and half female,which were randomly divided into a control group of 10,administered groups:the Strychnine Semen group,the processed Strychnine Semen group,the Brucine group,the Brucine N-oxide group,50 in each group,were given intragastric administration.The LD50 was determined by the Karber's method.During the period,the general state of the mice,their deaths,and the changes in body weights in the 14-day period after the administration of the four drugs were observed.HE staining was used to observe the pathological tissues damage of hearts,livers,spleens and kidneys in mice that died of poisoning.Results After administration by gavage,the poisoning reactions of the four drugs in mice were similar and showed a dose-dependent aggravation.There was no obvious abnormality in the body weight changes of the mice in each administration group compared with that of the blank control group within 14 days(P>0.05).The LD50 of processed Strychnine Semen was about 1.6 times of that of Strychnine Semen,the LD50 of Brucine could be up to 31 times of that of Brucine N-oxide.Compared with processed Strychnine Semen and Brucine N-oxide,the death-dose curves of Strychnine Semen and Brucine were steeper and dose-dependent.Compared with the blank control group,HE staining of hearts,livers,spleens and kidneys tissues of mice poisoned and killed in the highest dose group of the four drugs did not show obvious abnormality.Conclusion Processed Strychnine Semen and Brucine N-oxide are less toxic than Strychnine Semen and Brucine,which of treatment window are larger and safer.
