Action Mechanism of Jiaji EA in Treatment of CSR Based on Nrf2/ARE Signaling Pathway in Model Rats
Objective:To observe the effect of Jiaji electro-acupuncture(EA)in treatment of cervical spondylotic radiculopathy(CSR)in rats and to investigate its action mechanism in terms of Nrf2/ARE pathway.Methods:75 SPF-grade SD rats were randomly divided into the blank control group,the sham operation group,the model group,the acupuncture group and the EA group.The blank control group was not given any intervention,and the CSR model was established by surgically placing quantitative filter paper pieces soaked with formalin solution under the axilla of C8 nerve roots;whereas the sham operation group only exposed the nerve roots without placing filter paper pieces.The pain behavior,gait disturbance score and grasping ability score of rats in each group were observed after modeling.From the fourth day after modeling,the sham operation group and the model group were given daily binding for 15 minutes,the acupuncture group was given daily binding plus acupuncture for 15minutes,and the EA group was given daily binding plus EA for 15minutes.After the treatment,the serum content of MDA and the activities of SOD and GSH-PX were detected by kits,and the expressions of Nrf2,Keap1 and HO-1 in rat nerve root tissues were detected by Western blot technique.Results:The improvement degree and speed of pain behavior,gait disturbance and grasping ability in the acupuncture group and the EA group were more significant than those in the model group.Compared with those in the blank control group,there were no statistically significant differences in the serum levels of MDA and the activities of SOD and GSH-PX in the sham operation group(P>0.05).Compared with those in the sham operation group,the serum content of MDA was significantly higher,activities of SOD and GSH-PX were significantly lower,the expressions of Nrf2 and HO-1 were significantly decreased,and the expression of Keap1 was significantly increased in the model group(P<0.01).Compared with those in the model group,the serum content of MDA was decreased significantly(P<0.05),the activities of SOD and GSH-PX were increased significantly(P<0.05,P<0.01),the expressions of Nrf2 and HO-1 were significantly increased,and the expression of Keap1 was significantly decreased in the acupuncture group and the EA group(P<0.01).Compared with those in the acupuncture group,the serum content of MDA was significantly higher,the activities of SOD and GSH-PX were significantly lower(P<0.05),the expressions of Nrf2 and HO-1 were increased significantly(P<0.01,P<0.05),and the expression of Keap1 was decreased significantly in the EA group(P<0.01).Conclusion:Jiaji EA can improve gait impairment and reduce pain in CSR rats,and the mechanism may be related to the activation of Nrf2/ARE signaling pathway,up-regulation of Nrf2 and HO-1 expression and down-regulation of Keap1 expression.
