Impact of Electro-Acupuncture on Intestinal Barrier in Rats with T2DM
Objective:To observe the effects of electro-acupuncture on systemic inflammation and intestinal barrier function in rat model of type 2 diabetes mellitus(T2DM).Methods:SD rats were randomly divided into the normal group,the model group and the electro-acupuncture group,with 6 rats in each group.T2DM model was established by high-fat diet combined with single intraperitoneal injection of small-dose streptozotocin(STZ).The electro-acupuncture group was treated with electro-needling Zusanli(ST36),Sanyinjiao(SP6),Pishu(BL20)and Shenshu(BL23),3 times per week for 8 weeks.Fasting blood glucose(FBG)was measured after the treatment in each group.The serum contents of fasting insulin(FINS),tumor necrosis factor(TNF-α),interleukin-6(IL-6),lipopolysaccharide(LPS),D-lactic acid(D-LA)and diamine oxidase(DAO)were detected by ELISA.Homeostasis model assessment of insulin resistance(HOMA-IR)was calculated.Transmission electron microscopy was used to observe the ultrastructure of the small intestinal mucosal epithelium.WB was used to detect the expressions of the small intestinal epithelial tight junction proteins of ZO-1,Occludin and Claudin-1.Results:The levels of FBG,FINS and HOMA-IR were decreased(P<0.01),the serum inflammatory factors of TNF-α and IL-6 were reduced(P<0.01),the serum contents of LPS,D-LA and DAO were down-regulated(P<0.01),and the protein expressions of ZO-1,Occludin and Claudin-1 in the small intestine epithelium were up-regulated in the electro-acupuncture group than those in the model group(P<0.01).Conclusion:Electro-needling ST36,SP6,BL20 and BL23 can reduce FBG and alleviate systemic inflammation in T2DM rats,which is related to up-regulating small intestinal epithelial tight junction protein,improving intestinal mucosal permeability and repairing intestinal mucosal barrier function.
