针刺对卵巢储备功能减退大鼠血清激素及PI3K/Akt/mTOR信号通路蛋白表达的影响

目的:观察针刺对卵巢储备功能减退(DOR)大鼠血清激素、卵巢形态学变化及卵巢组织中p-PI3K、p-Akt及p-mTOR蛋白表达的影响,探索针刺治疗DOR的可能机制.方法:18只SPF级SD雌性大鼠随机分为空白组、模型组和针刺组,每组6只;模型组、针刺组采用腹腔注射环磷酰胺制备DOR大鼠模型,空白组仅腹腔注射同等量生理盐水.造模后针刺组取肾俞、关元、中极、气海、太溪、双侧三阴交和双侧子宫穴进行针刺,每次留针20 min,1次/d,连续21 d.空白组、模型组:仅抓取不进行干预,1次/d,连续21 d.观察比较各组间大鼠的血清激素改变情况,HE染色法观察大鼠卵巢形态学改变、卵巢中各级卵泡及黄体的数目变化,颗粒细胞层厚度改变;免疫组织化学法、Western blot法检测卵巢组织中p-PI3K、p-Akt及p-mTOR的蛋白表达.结果:与空白组比较,模型组血清FSH、LH水平升高(P<0.05),血清E2、AMH水平降低,差异具有统计学意义(P<0.05);卵巢组织中各级卵泡数明显减少,差异具有统计学意义(P<0.05);卵巢组织中PI3K、Akt及mTOR蛋白表达水平升高,差异具有统计学意义(P<0.05).与模型组比较,针刺组血清FSH、LH水平显著降低,差异具有统计学意义(P<0.05);血清E2、AMH水平显著升高,差异具有统计学意义(P<0.05);卵巢组织中各级卵泡数明显增多,差异具有统计学意义(P<0.05);卵巢组织中p-PI3K、p-Akt及p-mTOR蛋白表达水平降低,差异具有统计学意义(P<0.05).结论:针刺可以有效改善DOR大鼠卵巢储备功能,促进卵泡数量增加.作用机制可能与针刺调节卵巢组织中p-PI3K、p-Akt及p-mTOR蛋白表达有关.
Effect of Acupuncture on Expressions of Serum Hormones and PI3K/Akt/mTOR Signal Pathway in DOR Rats
Objective:To observe the effects of acupuncture on serum hormones,ovarian morphological changes,and protein expressions of p-PI3K,p-Akt and p-mTOR in ovarian tissues of rats with diminished ovarian reserve(DOR),and to explore the possible mechanism of acupuncture treatment for DOR.Methods:18 SPF grade SD female rats were randomly divided into the normal group,the model group and the acupuncture group,with 6 rats in each group.The DOR model was established by intraperitoneal injection of Cyclophosphamide.The normal group only received intraperitoneal injection of the same amount of normal saline.After modeling,the acupuncture group was treated with needling Shenshu(BL23),Guanyuan(RN4),Zhongji(RN3),Qihai(RN6),Taixi(KI3),bilateral Sanyinjiao(SP6),and bilateral Zigong(EX-CA1),with 20 minutes needle retention,once a day,a total of 21 times.The rats in the normal group and the model group were grabbed once a day for 21 days without any other intervention.The changes in serum hormones were compared among the groups.The morphological changes in the ovaries,the number of follicles and corpus luteum at different levels in the ovaries,and the thickness changes of the granulosa cell layer were observed by using HE staining method.The protein expressions of p-PI3K,p-Akt and p-mTOR in ovarian tissues were detected by immunohistochemical and Western blot.Results:Compared with those in the normal group,the serum levels of FSH and LH were increased(P<0.05),the serum levels of E2 and AMH were decreased(P<0.05),the number of follicles at all levels in ovarian tissues were significantly decreased(P<0.05),and the protein expressions of PI3K,Akt and mTOR in ovarian tissues were increased in the model group(P<0.05).Compared with those in the model group,the serum levels of FSH and LH were significantly decreased(P<0.05),the serum levels of E2 and AMH were significantly increased(P<0.05),the number of follicles at all levels in ovarian tissues were significantly increased(P<0.05),the protein expressions of p-PI3K,p-Akt and p-mTOR in ovarian tissues were decreased in the acupuncture group(P<0.05).Conclusion:Acupuncture can effectively improve ovarian reserve function and promote an increase in follicle count in DOR rats.The action mechanism may be related to regulating the protein expressions of p-PI3K,p-Akt and p-mTOR in ovarian tissues.
