Effects of Needling BL13,EX-B1 and RN17 on CHRM2 Receptor in Rats with Asthma
Objective:To investigate the effects of needling Feishu(BL13),Dingchuan(EX-B1)and Danzhong(RN17)on cholinergic receptor muscarinic 2(CHRM2)receptor in lung tissues of the rats with bronchial asthma.Methods:50 Wistar rats(6-8 weeks)were randomly divided into the normal control group, the model group, the acupuncture group, the BL13 group and the non-meridian and non-acupuncture group, with 10 rats in each group.Ovalbumin(OVA)was applied to sensitize and induce the model establishment of bronchial asthma.Lung function was detected by pulmonary function meter, pathological changes in lung tissues were observed by HE staining, changes in CHRM2 protein were detected by immunofluorescence, and CHRM2 protein content in lung tissues was determined by Western Blot. Results:Lung function test in rats showed that needling BL13,EX-B1 and RN17 could significantly reduce the increased airway resistance to reduce the airway inflammatory reaction in the rats with bronchial asthma.Pathology showed that bronchial inflammatory cell infiltration was increased in the model group.Immunofluorescence showed that the protein expressions of CHRM2 were significantly lower in the acupuncture group than those in the model group(P<0.01).Western Blot showed that the protein expressions of CHRM2 were significantly lower in the acupuncture group than those in the model group(P<0.01).Conclusion:Needling BL13,EX-B1 and RN17 can reduce the increased airway hyperresponsiveness in asthma rats by lowering CHRM2 levels.
