Effect of Moxibustion at ST36 on Gastric Motility and Mucin Expression in Gastric Mucosa of CAG Model Rats
Objective:To observe the effects of moxibustion at Zusanli(ST36)on gastric motility indexes,including gastric antral mobility index,gastric emptying rate and expressions of MUC1,MUC5AC and MUC6 in gastric mucosa of chronic atrophic gastritis(CAG)model rats.Methods:Healthy SD rats were randomly divided into the blank group(n=10)and the modeling group(n=120).The CAG model was established by compound etiological method.After successful modeling,the rats were randomly divided into the model group,the medication group and the moxibustion group,each group were subdivided into 4 subgroups according to 4 phases of 1 w,2 w,3 w and 4 w,with 10 rats in each subgroup.The blank group and the model group were not given any treatment.The moxibustion group was treated with mild moxibustion at ST36,30 min each time,once a day.The medication group was treated with Teprenone Solution,13.5 mg/kg,once a day for a month.In terms of gastric motility test,a color Doppler ultrasound diagnostic device was used to measure the antral relaxation area,the antral contraction area,and the total number of antral contractions within 3 min to calculate the antral mobility index.After intragastric administration for 30 min,the rats were euthanized to measure the gastric emptying rate.For mucin detection,the rats were euthanized at specific phase,and tissues were collected.immunohistochemistry and Western blotting were employed to detect the expression range and relative expression levels of MUC1,MUC5AC and MUC6.Results:In terms of the results of gastric motility:compared with those in the blank group,the gastric antral motility index and the gastric emptying rate were significantly down-regulated in the model group(P<0.01,P<0.05);compared with those in the model group during the same phase,the up-regulations of the gastric antral motility index were statistically significant in the medication group and the moxibustion group at 2 w,3 w and 4 w(P<0.05);compared with those in the medication group during the same phase,the up-regulations of gastric antral motility index were statistically significant in the moxibustion group at 2 w,3 w and 4w(P<0.05).In terms of immunohistochemical examination:compared with the those in the blank group,the expressions of MUC1,MUC5AC and MUC6 were down-regulated in the model group(P<0.05);compared with that in the model group during the same phase,the expression of MUC1 was up-regulated in the moxibustion group and the medication group(P<0.05);compared with that in the simultaneous medication group,the expression of MUC1 was up-regulated in the moxibustion group at 3 w and 4 w(P<0.05).In terms of Western blotting results:the relative expressions of MUC1,MUC5AC and MUC6 were down-regulated in the model group compared with those in the blank group(P<0.05);compared with that in the simultaneous phase model group,the relative expression of MUC1 was up-regulated in the moxibustion group and the medication group(P<0.05);compared with that in the simultaneous phase medication group,the relative expression of MUC1 was up-regulated in the moxibustion group at 4 w(P<0.05).Conclusion:As a protective agent of gastric mucosa,Teprenone can promote the repair of gastric mucosa in rats,but it shows no effect on the gastric motility.Moxibustion at ST36 can not only promote the repair of gastric mucosa in rats,but also promote the gastric motility.However,moxibustion takes effect relatively late,at least two weeks of continuous moxibustion are needed for gradual effectiveness,and based on the current experimental results,the longer the moxibustion duration,the better the therapeutic effect.
