Exploring Acupuncture's Mechanism in Treating PDC Based on Bioinformatics and Network Topology Analysis
Objective:To investigate the mechanism of acupuncture in treatment of Parkinson's disease-related constipation(PDC).Methods:The literature on acupuncture treating PDC over the past 10 years was reviewed.Active ingredients that exhibited significant changes before and after the treatment were identified,and their related targets were retrieved from PubChem and Swiss Target Prediction database.The intersection of active ingredients and disease targets was determined,the PPI network was constructed,and the molecular functions,cell components,biological processes and pathways of proteins involved were obtained by enrichment analysis of GO function and KEGG signaling pathways of key therapeutic targets through the Bioinformatics platform.Results:Predictive analysis identified 171 gene targets associated with acupuncture treatment for PDC,and revealed 10 KEGG enrichment analysis pathways.Conclusion:Acupuncture may regulate DA,α-Syn,NE and 5-HT levels through core targets of DRD2,GRM5,SLC6A3,MAOB and HTR1A in treatment of PDC,the main signaling pathway calcium signaling pathway,G protein-coupled receptor signaling pathway,and neuroactive ligand-receptor interaction.
