Effects of Invigorating Qi and Activating Blood EA on Hippocampal HMGB1 Inflammatory Pathway in Rats with CSR
Objective:To investigate the therapeutic effect of invigorating Qi and activating blood acupuncture on radicular pain and the central anti-inflammatory and analgesic mechanism in rats with cervical spondylotic radiculopathy(CSR).Methods:The male SD rats were randomly divided into the blank control group,the sham operation group,the model group and the electro-acupuncture(EA)group,with 6 rats in each group.The CSR model was established by the method of vertebral tube insertion,and the intervention started three days after modeling.The EA group was treated with electro-needling Jingjiaji(EX-B2),Zusanli(ST36)and Geshu(BL17),with the intensity of 1.5mA,frequency 2/50 Hz,30min/times,once a day for 2 weeks.Behavioral evaluations were conducted on the first day before the intervention,and the 3rd,5th,7th,10th,and 14th day after the intervention start.Pathological changes of the left nerve root were observed by HE staining.The contents of IL-1β and TNF-α in the hippocampus were detected by ELISA.The protein expressions of HMGB1,TLR4 and NF-κB in the hippocampus were detected by Western Blot.Results:Compared with those at each time point in the sham operation group,the paw withdrawal thermal latency(PWTL)and the paw withdrawal mechanical threshold(PWMT)were significantly decreased in the model group,and the nerve root tissue damage was more serious,with part of the neuronal cells ruptured and deformed,severe cell edema,disorganized fiber arrangement,part of the fibers broken,and cytosolic bodies reduced;the contents of IL-1β and TNF-α in the hippocampus were significantly increased(P<0.01),and the protein expressions of HMGB1,TLR4 and NF-κB were significantly up-regulated in the model group(P<0.01).Compared with those in the model group,the PWTL was significantly increased from the 5th day of the intervention,and the PWMT was significantly increased from the 7th day of the intervention in the EA group,with a significant reduction in the damage to the nerve root tissues,and significant decreases in the contents of hippocampal IL-1β and TNF-α(P<0.01);the protein expressions of HMGB1,TLR4 and NF-κB were significantly down-regulated in the EA group(P<0.01).Conclusion:Invigorating Qi and activating blood EA can alleviate radicular pain in CSR rats,and its mechanism may be related to the inhibition of hippocampal inflammation through HMGB1 signaling pathway.
