为了鉴定市场上繁多的羊肚菌品种,了解羊肚菌栽培品种间的遗传多样性水平及亲缘关系.研究选用了41份国内市场上的栽培菌株,运用多基因联合矩阵序列对羊肚菌种类进行甄别,同时利用ISSR分子标记技术结合形态学指标进行遗传多样性分析.结果表明:基于多基因序列的系统发育树将 41份菌株聚为两大支,39份来自不同地区的菌株与六妹羊肚菌Morchella sextelata聚为一支,其余 2份菌株与梯棱羊肚菌Morchella importu-na聚为一支.10条ISSR引物扩增得到 62个条带,多态性比例达 85.48%,有效等位基因数(Ne)为 1.372 2个,基因多样性指数(H)为 0.241 3,Shannon's 信息指数(I)为 0.386 7,不同地理来源的群体间的遗传分化系数和遗传距离与地理位置的远近没有明确相关性,主成分分析结果也表明与地理位置相关性不明显,群体结构分析中有37份材料的Q值≥0.6,大部分羊肚菌材料血缘比较单一;基于表型性状和ISSR的聚类结果均可将供试材料分为5组,部分材料的形态学标记与ISSR标记聚类结果存在一定差异.本研究通过对不同来源的羊肚菌菌株进行分子鉴定和遗传多样性分析,为羊肚菌菌种资源的鉴定、开发及利用提供一定的理论参考依据.
Identification and Genetic Diversity of Cultivated Morchella spp.
