不同种源秋茄树幼苗生长和抗寒生理差异分析

低温是限制秋茄树Kandelia obovata向北分布的主要因子,浙江省是秋茄树分布的最北界,因此筛选抗寒种质,对红树林北移引种栽培具有重要意义.本研究以福建漳江口、福建福鼎和浙江龙港 3个种源的秋茄树胚轴为材料,通过室内栽培试验,调查不同种源秋茄树幼苗的生长特性,并利用人工气候培养箱,设置了18℃、4℃和0℃共 3个温度,每个温度处理 24 h后,观察幼苗受寒害情况,测量分析不同种源秋茄叶片生理生化指标变化规律.结果表明:福建漳江口秋茄树幼苗的株高、地径生长量以及生物量均显著高于其他 2个种源;低温胁迫下,相较于其他 2个种源,福建漳江口种源秋茄树幼苗受寒害最为严重;低温胁迫下各种源幼苗的渗透调节物质含量总体呈上升趋势,在 0℃时福建福鼎种源幼苗的渗透调节物质含量显著高于福建漳江口种源和浙江龙港种源,分别高出 147.4%和 84.0%;随着温度的下降,秋茄树幼苗叶片的SOD活性福建漳江口种源呈先上升后下降趋势,福建福鼎种源呈先下降后上升趋势,而浙江龙港种源呈上升趋势,在 0℃时,浙江龙港种源秋茄树幼苗的SOD活性最高;低温胁迫下各种源秋茄树幼苗的MDA含量均呈上升趋势,在 0℃时,福建漳江口种源幼苗的MDA含量显著高于福建福鼎种源和浙江龙港种源,分别高出 117.1%和 70.0%.因此,在相同条件下,浙江龙港种源秋茄树幼苗的生长量相对较低,但随着低温胁迫程度的增加表现出较强的PRO和SOD活性以及可溶性糖含量的上升,抗寒能力最强.在浙江开展红树林营建,建议优先选择浙江龙港种源秋茄树幼苗.本研究为红树植物秋茄树的北移引种栽培和生态保护提供了重要参考依据.
Variance of Growth and Physiological Difference in Cold Resistance of Kandelia obovate Seedlings from Different Provenances
Low temperature is the predominating factor for Kandelia obovata northward distribution,screening cold-resistant germplasm is of great significance for mangrove northward migration,introduction cultivation.This study provides the important reference for the northward introduction cultivation of mangrove plant K.obovata and the ecological conservation.The hypocotyls of three provenances from Zhangjiang Estuary,Fuding in Fujian Province and Longgang in Zhejiang Province were used as objects,the growth characteristics of K.obovata seedlings were investigated through indoor cultivation experiments.The cold damage of seedlings was observed after each temperature was treated for 24 hours with the artificial climate incubators used to set temperatures of 18℃,4℃,and 0℃,analyzing changes in physiological and biochemical indexes of the leaves from different provenances.The results showed that the plant height,ground diameter growth,biomass of the seedlings in Zhangjiang Estuary were significantly high-er than those from other provenances.Under the stress of low temperature,the seedlings in Zhangjiang Estuary of Fujian Province suffered the most severe cold damage compared with the other two provenances.At 0℃,the content of osmoregulatory species in Fuding provenance was significantly higher than those in Zhangjiang Estuary and Longgang provenance,with 147.4%and 84.0%,respectively.With the descent of temperature,the SOD activity in the leaves of Zhangjiang Estuary seedlings in Fujian Province increased first and then reduced,the SOD activity in Fuding seedlings de-creased first and then increased,and the SOD activity in Longgang seedlings increased with the highest at 0℃.The MDA content of the seedlings from Zhangjiang Estuary in Fujian Province was significantly higher than those from Fu jian Province and Zhejiang Province,which was 117.1%and 70.0%higher at 0℃,respectively.Therefore,under the same conditions the seedling growth of Longgang provenance in Zhejiang Province was rela-tively low;however,with the increase of low-temperature stress,the seedlings showed strong PRO and SOD activities and increase of soluble sugar content,displaying the strongest cold resistance,accordingly,K.obovata seedlings of Longgang provenance should be prioritized in the mangrove con-struction.
