Analysis on Accumulation of Cadmium and Manganese in Brown Rice of Derivatives from Two Nramp5 Gene Mutants of Rice in Cadmium-contaminated Paddy Field
The derivatives of deletion mutation of Nramp5 gene from Luohong 4A,point mutation of Nramp5 gene from lcdl and heterozygotes of deletion and point mutation of Nramp5 gene in rice were used to characterized the contents of cadmium(Cd)and manganese(Mn)in brown rice,which were of different genetic backgrounds and planted in cadmium-contaminated paddy fields(0.58 mg/kg available Cd at pH 5.3).The results showed that the genetic background did not affect the low accumulation characteristics of Cd and Mn in brown rice caused by deletion or point mutation of Nramp5 gene.The average Cd content in brown rice with heterozygotes of deletion and point mutation of Nramp5 gene was 0.082 mg/kg,which was significantly lower than the control variety 9311 containing wild-type Nramp5 gene(2.345 mg/kg),and there was no significant difference in Cd and Mn content compared with that of the homozygotes of deletion or point mutation of Nramp5 gene,respectively.All of the Cd contents in brown rice of deletion mutation homozygote,point mutation homozygote and heterozygote of deletion and point mutation were lower than the threshold of the national food safety standard(≤0.2 mg/kg).The results indicated that the hybrid still had the characteristics of low Cd accumulation in brown rice even though one parental line was of Nramp5 gene deletion mutation from Luohong 4A and the other parental line was of Nramp5 gene point mutation from lcd1,which provides important reference data for molecular breeding of low Cd accumulation in hybrid rice.
