Physiological Changes and Transcriptome Analysis of Glume-gapping in Rice Male Sterile Lines
To explore the mechanism of glume-gapping in rice male sterile lines and identify the key genes of glume-gapping in rice,hoping to reduce the occurrence of glume-gapping in hybrid rice seed production,the male sterile lines Xiangyu 5A(XY5A)and Yue 020S(Y020S)with significant differences in glume-gapping traits,and their restorer line R3576 were used as experimental materials.The glumes of XY5A and Y020S at flowering stage were collected for transcriptome sequencing analysis,and the physiological indexes related to glume opening and closing were determined.The results showed that the contents of soluble sugar and jasmonic acid in XY5A with heavily glume-gapping were higher than those of Y020S with lightly glume-gapping,while the content of auxin was exactly the opposite.Transcriptome analysis obtained 3 099 differentially expressed genes(DEGs).GO enrichment analysis showed that these DEGs mainly play a role in metabolic and catalytic processes.KEGG Pathway analysis showed that these DEGs were mainly enriched through plant-pathogen interaction,phytohormone signal transduction,starch and sucrose metabolism.Eleven genes that might be closely related to glume-gapping occurrence were screened from starch and sucrose metabolic pathways,phytohormone signal transduction pathways and transcription factors.The results indicated that the genes such as Os07g0543300,OsJARl and OsMADS16 might be the key genes leading to glume-gapping in rice male sterile lines.In this study,the differentially expressed genes and their functional annotation information of rice male sterile lines with different glume-gapping traits were obtained,laying a foundation for further exploring the molecular mechanism of glume-gapping in rice male sterile lines.
