美洲大蠊研粉对脊髓损伤大鼠氧化应激的影响

目的 探讨美洲大蠊研粉对脊髓半横断损伤(hSCI)大鼠脊髓损伤区域氧化应激的影响。方法 60 只雌性SD大鼠按照随机数字表法分为对照组和观察组,各 30 只,两组均通过打开T10椎板行左侧脊髓半横断,制作hSCI模型。术前 30 min及术后 1~14 d,对照组予 0。9%氯化钠溶液、观察组予美洲大蠊研粉(630 mg/kg)溶于 0。9%氯化钠溶液灌胃,5 mL/只,每天 1 次。术后 6h及 1、3、7、14 d每组每个时间点取 5 只大鼠,用酶联免疫吸附试验检测丙二醛(MDA)、8-羟基脱氧鸟苷(8-OHdG)含量;术后 14d每组取 5 只大鼠用于脊髓组织尼氏染色。结果 与对照组比较,观察组在显微镜10×20倍及10×40倍视野下脊髓组织神经元数目均更多[(26。25±0。50)个比(10。84±0。58)个、(14。92±0。32)个比(6。75±0。32)个,P<0。01];观察组术后 7、14 d脊髓组织MDA[(0。61±0。03)nmol/mL比(0。89±0。02)nmol/mL、(0。54±0。01)nmol/mL比(0。68±0。02)nmol/mL,P<0。01]和 8-OHdG[(17。92±0。99)ng/mL比(23。37±0。77)ng/mL、(15。91±0。28)ng/mL比(20。18±0。37)ng/mL,P<0。01]含量均低于对照组;与对照组比较,观察组氧化应激高峰时间更早,MDA和 8-OHdG表达的峰值提前到了术后 3d。结论 美洲大蠊研粉降低了hSCI大鼠脊髓损伤区域MDA、8-OHdG的表达,使氧化应激高峰时间提前,减轻了氧化应激损伤,减少了神经元丢失。
Effect of Periplaneta Americana powder on oxidative stress in rats with spinal cord injury
Objective To investigate the effect of Periplaneta Americana powder on oxidative stress in the spinal cord injury area of rats with hemi-transected spinal cord injury(hSCI).Methods Sixty female SD rats were randomly divided into a control group and an observation group,with 30 rats in each group.The hSCI model was established in both groups by opening the T10 vertebral plate to perform left spinal cord hemisection.Thirty min-utes before surgery and 1~14 days after surgery,the control group was administered 0.9%sodium chloride solution,while the observation group was given Periplaneta Americana powder(630 mg/kg)dissolved in 0.9%sodium chloride solution via gavage,5 mL per rat,once daily.Five rats were taken from each group at each time point 6 hours and 1,3,7,and 14 days after surgery,and the malondialdehyde(MDA)and 8-hydroxydeoxyguanosine(8-OHdG)contents were detected by enzyme-linked immunosorbent assay.Five rats were taken from each group 14 d after surgery for Nissl staining of spinal cord tissue.Results Compared with the control group,the observation group showed a significantly greater number of neurons in the spinal cord tissue under the microscope field of view of 10×20 and 10×40[(26.25±0.50)vs.(10.84±0.58),(14.92±0.32)vs.(6.75±0.32),P<0.01].The observation group had lower levels of MDA[(0.61±0.03)nmol/mL vs.(0.89±0.02)nmol/mL,(0.54±0.01)nmol/mL vs.(0.68±0.02)nmol/mL,P<0.01]and 8-OHdG[(17.92±0.99)ng/mL vs.(23.37±0.77)ng/mL,(15.91±0.28)ng/mL vs.(20.18±0.37)ng/mL,P<0.01]in the spinal cord tissues at days 7 and 14 after surgery.Compared with the control group,the peak time of ox-idative stress in the observation group occurred earlier,and the peak values of MDA and 8-OHdG expression were advanced to 3 days after surgery.Conclusion Periplaneta Americana powder reduced the expression of MDA and 8-OHdG in the spinal cord injury area of hSCI rats,advanced the peak time of oxidative stress,alleviated oxida-tive stress damage,and reduced neuronal loss.
