摘要
泛素特异性肽酶(USP)是去泛素化酶家族的主要成员,其失调可能会导致泛素系统功能障碍,进而调控肿瘤的进展.USP通过选择性地降解或稳定癌基因、抑癌基因、DNA损伤修复和细胞周期等相关的底物蛋白来调节肝细胞癌(HCC)的发生、发展.文章总结关于USP在HCC中的研究进展及USP特异性小分子抑制剂的应用,为通过靶向USP诊断和治疗HCC提供参考依据.
Abstract
Ubiquitin-specific protease(USP)is a major member of the deubiquitinating enzymes(DUB)family.Dysregulation of DUB could lead to dysfunction of the ubiquitin system,which in turn regulates the tumor progression.USP regulates the development and progression of hepatocellular carcinoma(HCC)by selectively degrading or stabilizing substrate proteins related to oncogenes,tumor suppressors,DNA damage repair,and cell cycle.This article summarizes the research progress of USP in HCC and the application of USP-specific small molecule inhibitors,providing a reference for targeting USP as a diagnostic and therapeutic tool for HCC.
