摘要
目的 比较3种工艺条件下所得地黄当归胶囊对更年期阴虚内热证模型大鼠的影响,以筛选最佳工艺.方法 通过手术去除双侧卵巢,然后连续灌服热性中药(制附子25 g,干姜25 g,加水煎煮成100 mL,质量浓度为50%的制剂),建造大鼠更年期黄体生成素阴虚内热证模型,以大鼠给药前后体质量、肛温的变化,以及末次给药后的子宫系数,血清中血清促卵泡激素水平(FSH)、血清黄体生成素(LH)、雌二醇(E2)水平为指标,对不同工艺所得地黄当归胶囊进行筛选.结果 与模型组相比较,地黄当归胶囊A工艺高、低剂量组,B工艺高、低剂量组大鼠体质量增长较快(P<0.05或P<0.01),B工艺高、低剂量组肛温下降较多(P<0.05或P<0.01);A工艺高剂量组,B工艺高、低剂量组可明显增加大鼠子宫系数(P<0.01);A工艺高剂量组,B工艺高、低剂量组明显降低FSH水平(P<0.01);B工艺高剂量组明显降低LH含量(P<0.05);B工艺高、低剂量组能明显升高大鼠血清中E2水平(P< 0.05或P< 0.01).结论 在相同实验条件下,地黄当归胶囊B工艺所得提取物对更年期阴虚内热证模型大鼠有较明显的疗效.
Abstract
Objective To screen the optimum processing for Dihuang Danggui capsules by the comparison of the impact on climacteric deficiency heat syndrome rat model.Methods Rat climacteric deficiency heat syndrome models of menopause were induced by the removal of bilateral ovaries.The rats were fed with the hot medicine everyday,namely Aconite 25 g and Ginger 25 g,adding water to 100 mL decoction,until reaching 50% concentration.The body weight,rectal temperature changes of before and after the administration,the uterine factor and rat serum follicle stimulating hormone (FSH),luteinizing hormone (LH),and estradiol (E2) levels after the last administration were used as indicators of different processes for Dihuang Danggui capsules to be screened.Results Compared with the model group,weight gain was observed in both Dihuang Danggui capsule A (high and low dose groups)and capsule B (high and low dose groups) (P < 0.05 or P < 0.01);the rectal temperature was decreased in capsule B high and low dose groups (P < 0.05 or P < 0.01);the uterine factor was increaseed in A high dose group and B high and low dose groups (P <0.01);the level in FSH of A high dose group and B high and low dose group was decreased;the level of LH in B high dose group was decreased (P < 0.05);the level of E2 in B high and low dose groups was increased (P < 0.05 or P < 0.01).Conclusion Under the same condition,extract of Dihuang Danggui capsule B has obvious cure effect on climacteric deficiency heat syndrome rat model.
