HPLC determination of the content of hydroxy safflower yellow A and ferulic acid in Dieda Huoxue tincture
Objective To establish an HPLC method to determine the content of hydroxy safflower yellow A and ferulic acid in Dieda Huoxue tincture.Methods ODS2 C18 (4.6 mm× 150 mm,5 μm) was used as chromatographic column.Chromatographic conditions for hydroxy safflower yellow A:methanol-0.7% phosphoric acid solution (25:75) as the mobile phase,with the flow rate 1 mL · min-1,detection wavelength 403 nm,column temperature 30 ℃,and injection volume 10 μL.Chromatographic conditions for ferulic acid:acetonitrile-0.085% phosphoric acid solution (17:83) as the mobile phase,with the flow rate 1 mL · min-1,detection wavelength 316 nm,column temperature 35 ℃,and injection volume 10 μL.Results Hydroxy safflower yellow A had a good linearity at 0.0478-2.39 μg (r2 =0.9996),and the average recovery was 98.6%.Ferulic acid had a good linearity at 0.028 35-2.835 μg (r2 =0.9998),and the average recovery was 99.1%.Conclusion The method is simple,accurate,and highly sensitive,and can be used for the quality control of Dieda Huoxue tincture.
