STAT3 inhibitor niclosamide improves the cognitive impairment in mice with Alzheimer's disease and related mechanism
Objective To determine the effect and mechanism of signal transducers and activators of transcription 3(STAT3)in the occurrence and development of cognitive impairment in mice with Alzheimer's disease.Methods Alzheimer's disease model was established by stereotaxic injection ofβ-amyloid protein into the bilateral hippocampus of C57BL/6J mice.Then niclosamide,an inhibitor of STAT3 was given.Animal behavior analysis and Western blot were used to determine the effect and mechanism of STAT3 in the cognitive impairment.Results The behavior test showed that compared with the model group,the anxiety symptoms,spatial exploration ability,object recognition ability and learning and memory were improved ability in the treatment group.Western blot showed that the pathological markers of Alzheimer's disease were improved and the expression of pro-inflammatory factors was down-regulated.The oxidative stress related indexes of the treatment group were improved by kit analysis.HE staining showed that the histomorphology of the hippocampal neurons in the treatment group was improved.Conclusion It is preliminarily proved that inhibition of STAT3 can reduce the neuroinflammation and oxidative stress and improve the cognitive impairment in mice with Alzheimer's disease.
