川枳壳黄酮类成分及其对成骨细胞增殖作用的研究

目的 研究川枳壳黄酮类化学成分及其对成骨细胞增殖的影响.方法 采用聚酰胺树脂、葡聚糖凝胶、硅胶柱色谱等方法进行成分分离,通过质谱和核磁共振波谱分析与鉴定结构,采用体外细胞实验对所得化合物进行成骨细胞增殖活性检测.结果 从川枳壳中分离得到12个黄酮类及其苷类成分,分别鉴定为柚皮苷(1)、新橙皮苷(2)、柚皮素-7-O-β-D-葡萄糖苷(3)、narirutin(4)、sarotanoside(5)、圣草酚-7-O-β-D-葡萄糖苷(6)、柚皮素(7)、芜花素(8)、圣草酚(9)、angophorol(10)、南酸枣苷(11)、tetrahydroprotogenkwanone(12).其中化合物 1~4、6~9、12具有促进成骨细胞增殖作用,且化合物12的细胞增殖率达196.3%,优于阳性药雌二醇.结论 化合物5、8、10、11、12为首次从枳壳中分离得到,化合物5、10、12为首次从柑橘属中分离得到,化合物1~4、6~9、12对成骨细胞具有明显的增殖作用.
Flavonoids from Sichuan Aurantii Fructus and their effects on the proliferation of osteoblasts
Objective To determine the chemical constituents of flavonoids from Sichuan Aurantii Fructus and their effects on the proliferation of osteoblasts.Methods Polyamide resin,dextran gel,silica gel column chromatography and other methods were used to separate the components.The structures were analyzed and identified by mass spectrometry and nuclear magnetic resonance spectroscopy.The obtained compounds were tested for osteoblast proliferation activity by in vitro cell experiments.Results Twelve flavonoids and their glycosides were isolated from Sichuan Aurantii Fructus,and were identified as naringin(1),neohesperidin(2),naringenin-7-O-β-D-glucopyranoside(3),narirutin(4),sarotanoside(5),eriodictyol-7-O-β-D-glucopyranoside(6),naringenin(7),genkwanin(8),eriodictyol(9),angophorol(10),choerospondin(11),and tetrahydroprotogenkwanone(12).Compounds 1~4,6~9 and 12 promoted osteoblast proliferation,and the cell proliferation rate of compound 12 reached 196.3%,which was superior to that of estradiol.Conclusion Compounds 5,8,10,11,and 12 have been isolated from Aurantii Fructus for the first time,while 5,10,and 12 from Citrus for the first time.Compounds 1~4,6~9 and 12 obviously promote the proliferation of osteoblasts.
