Effect of Gegen Qinlian decoction combined with probiotics on intestinal microbiota and related inflammatory factors in mice with ulcerative colitis
Objective To determine the effect of Gegen Qinlian decoction(GQD)combined with probiotics on the gut microbiota and related inflammatory factors in mice with ulcerative colitis.Methods Totally 45 male C57BL/6 mice were randomly divided into 7 groups:a Normal group,a Model group,a Mes group(0.6 g·kg-1),a Probiotics group(0.3 g·kg-1),a GQD L+Probiotics group,a GQD M+Probiotics group,a GQD H+Probiotics group(2.54,5.08,and 10.16 g·kg-1).Except for the Normal group,all other groups were subjected to 7 continuous days of free drinking of 3%dextran sodium sulfate solution(DSS),followed by 7 days of drinking water before the modeling.The entire experiment was repeated 4 cycles.On the 35th day of modeling,mice with failed modeling were excluded based on the disease activity index(DAI),and the remaining mice were administered in groups for 21 days.The weight and DAI of mice during the modeling and administration period were recorded twice a week.After the experiment,the length of the colon was recorded,the pathological changes of the colon tissue were observed with HE staining,and colon mucosa damage index(CMDI)was scored.The expression of IL-6,TNF-α,and IL-10 in the serum was detected with ELISA.The diversity of intestinal microbiota in the mice through 16S rRNA sequencing of intestinal feces was analyzed.Results Compared with the Model group,in each treatment group,the body weight of mice were increased,the DAI were decreased,and the colon lengths were recovered.The recovery effect of the GQD M+Probiotics group,and GQD H+Probiotics group was significant(P<0.05),in a dose dependent manner.The expression level of pro-inflammatory factors IL-6 and TNF-α was significantly downregulated(P<0.01),with the expression level of IL-10 was significantly increased(P<0.01)in the combination therapy groups,indicating a dose-dependent effect.Alpha diversity and beta diversity of the gut microbiota showed that the species composition and species richness had changed among the different treatment groups.A decrease in species abundance in the Model group was found,while the Probiotics group and the GQD M+Probiotics group significantly restored species richness(P<0.01).Based on the phylum level,the dominant gut microbiota in the mice included Firmicutes and Bacteroidota,and the level of F/B was significantly upregulated in the combined administration group.Based on the species relative abundance at the genus level,beneficial bacteria such as Muribaceae,Lachnospiraceae NK4A136_group,Alistipes,Roseburia,and Lactobacillus were significantly reversed in the combined administration group.The conditional pathogenic bacteria such as Bacteroides,Colidextribacter and Clostridia_UCG-014 were significantly decreased.The intestinal flora were regulated and restored to steady state in the combined administration group significantly(P<0.05).Conclusion Combination of Gegen Qinlian decoction and probiotics has a positive effect on improving the intestinal microbiota imbalance,restoring the colon length,repairing pathological damage to the colon,and inhibiting the expression of pro-inflammatory factors in mice with ulcerative colitis.
