Effect of culture medium additives on the production and glycosylation of bispecific antibodies
Objective To determine the effect of culture medium additives(manganese ion,zinc ion,and sodium butyrate)on the growth,antibody production and critical quality attributes of Chinese hamster ovary(CHO)cells expressing bispecific antibodies.Methods Response surface design and center composite design were performed using JMP 13.0 software,to determine the effect of medium additives on growth,antibody production and Man5 levels.Results Additions of 1000 nmol·L-1 manganese chloride,30 μmol·L-1 zinc sulfate and 250 μmol·L-1 sodium butyrate in the media increased the peak viable cell density by 11.8%,increased the antibody production by 14.5%,but decreased the Man5 level by 28.9%.Conclusion Manganese ion,zinc ion and sodium butyrate play key roles in improving the bispecific antibody production and reducing the Man5 content.
