Fingerprint and content determination of Mongolian medicine hypecoum formula granules
Objective To establish HPLC fingerprint and content determination method of its active ingredients in Mongolian medicine hypecoum formula granules,and to provide a reference for the quality control of hypecoum formula granules.Methods An COSMOSIL 5 C18 column(4.6 mm×250 mm,5 μm)was used with the mobile phase consisting of acetonitrile-0.1 mol·L-1 ammonium acetate(containing 0.05%glacial acetic acid)for gradient elution.The flow rate was 1.0 mL·min-1,the column temperature was 28℃,the detection wavelength was 300 nm,and the injection volume was 10 μL.The fingerprint of hypecoum formula granules was established and the content of the 3 identified chemical ingredients was determined.Results The HPLC fingerprint of hypecoum formula granules was established,and 18 common peaks were identified.The similarity of 15 batches of hypecoum formula granules ranged from 0.982 to 1.000.The content of rutin,protopine,and coptisine in 15 batches of hypecoum formula granules ranged from 0.12%to 0.23%;0.50%to 0.54%;and 0.04%to 0.05%.Conclusion The method is stable and accurate,with simple operation and good repeatability,providing a reference for the evaluation and quality control of hypecoum formula granules.
