Chemical constituents in three origins of Rabdosia serra and their administration serum analysis in hepatic fibrosis mice based on UHPLC-Q-Exactive Orbitrap MS
Objective To identify the serum chemical compositions in three origins of Rabdosia serra and the serum in mice with hepatic fibrosis after the oral administration by UHPLC-Q-Exactive Orbitrap-MS.Methods The three origins of Rabdosia serra and the serum samples administered in mice were collected and analyzed with the chromatographic column of Shim-pack GIST-HP C18(2.1 mm×150 mm,3 µm).The mobile phase was 0.1%formic acid water(A)-methanol(B)for gradient elution and the flow rate was 0.3 mL·min-1.The column temperature was 30℃.The chemical compositions in three origins of Rabdosia serra and the serum in mice after the oral administration were analyzed with the high resolution mass spectrometry data and mass spectrometry fragmentation law of the compound.Results Totally 40 compounds were identified in the three origins of Rabdosia serra,mainly diterpenoids,flavonoids and phenolic acids.Two blood components and two possible metabolites were analyzed in the serum of mice.Conclusion This study preliminarily identifies the blood components in three origins of Rabdosia serra in mice with carbon tetrachloride induced liver fibrosis,providing a theoretical reference for the efficacy in the treatment of liver fibrosis.
